Objective:This study was to evaluate the ratio of Th17/Treg cells and their specific transcription factors in human peripheral blood lymphocytes(hPBLs)of patients with chronic periodontitis(CP)or aggressive periodontitis(AgP),and to investigate the association between AgP and imbalance of Th17/Treg in hPBLs,and the effect of periodontal treatment on Th17/Treg cells.METHODS:15 ml peripheral blood was drawn from each subject.The lymphocytes were isolated and divided into 2 samples that comprised 2/3 and 1/3 of the lymphocytes respectively.The ratio of Th17/Treg was evaluated using the first sample by flow cytometry,Realtime-PCR method was applied to access ratio the specific transcription factors,RORyt and FoxP3 using the second sample.SPSS13.0 software package was used to analyze the data.RESULTS:A total of 65 subjects were included in this study,which consists of 15 healthy control,25 severe periodontitis patients,and 25 AgP patients.The ratios of Th17/Treg and RORyt/FoxP3 in the subjects with AgP was significantly higher than those in the healthy controls(p<0.0001)and subjects with severe CP(p<0.0001).The ratios of Th17/Treg and RORyt/FoxP3 were significantly lower in those who had their periodontal inflammation controlled compared with those whose periodontal inflammation existedCONCLUSIONSThis study implys that the imbalance of Th17 and Treg cells exists in CP and AgP patients' peripheral blood lymphocytes and associates with the occurrence and development of these diseases.
