Objective To detect the expression of RbAP48 and E2F1 in oral squamous cell carcinoma,and to analyze the relationship between them and clinicopathological parameters,and to explore its clinical significance.Methods The expressions of RbAP48 and E2F1 in 70 cases of OSCC were detected by immunohistochemistry.The correlation between the expression of RbAP48and E2F1 was analyzed by chi-square test(?~2 test).The correlation between the two was analyzed by Spearman rank correlation analysis.law.Results(1)The results of immunohistochemical examination of 70 cases of oral squamous cell carcinoma showed that RbAP48 was mainly expressed in the nucleus of OSCC tissue,showing a yellow or brown-yellow granular distribution,and its positive expression rate was 64.29%(45/70);E2F1 is mainly expressed in the cytoplasm and nucleus of OSCC tissue,with a diffuse distribution of yellow or brown-yellow granules,and the cell nucleus is deeply stained with brownish yellow.The positive expression rate of E2F1 is 55.71%(39/70).(2)The positive expression rate of RbAP48 in the well-differentiated group was 73.68%(14/19),and the positive rate in the poorly-differentiated group was 60.78%(31/51).There was no significant difference between the two groups(P>0.05);the positive expression rate of RbAP48 in I+II group was 44.83%(13/29);in group III+IV,it was 78.05%(32/41).The difference between the two groups was statistically significant(P<0.05).The positive expression rate of RbAP48 in lymph node metastasis positive group was 82.35%(28/34),and that in lymph node metastasis negative group was 47.22%(17/36).There was significant difference between the two groups(P<0.05).The results showed that the expression of RbAP48 in OSCC was associated with clinical stage and lymph node metastasis,and was not significantly correlated with differentiation,age and gender.(3)The positive expression rate of E2F1 in the OSCC highly differentiated group was 52.63%(10/19),and that in the moderately and poorly differentiated group was 56.86%(29/51).There was no significant difference between the two groups(P>0.05).In the clinical stage,the positive expression rate of E2F1 in the I+II group was 55.17%(16/29);in the III+IV group was 56.10%(23/41),there was no significant difference between the two groups(P>0.05).The positive expression rate of E2F1 in the lymph node metastasis positive group was 50.00%(17/34),and the lymph node metastasis negative group was 61.11(22/36).There was no significant difference between the two groups(P>0.05).Statistical analysis showed that there was no significant correlation between E2F1expression in OSCC and clinical pathological parameters(lymph node metastasis,clinical stage,degree of differentiation,patient gender and age).(4)In 70 cases of oral squamous cell carcinoma,RbAP48 and E2F1 were positively expressed in 26 cases,RbAP48 and E2F1 were negatively expressed in 19 cases,RbAP48 was positive and E2F1 was negative in 12 cases,RbAP48 was negative and E2F1 was negative.There were 13 cases that were positive.By Spearman rank correlation analysis,there was no significant correlation between RbAP48 and E2F1 in oral squamous cell carcinoma(rs=0.143,P>0.05).Conclusions This study confirmed:(1)RbAP48 expression in oral squamous cell carcinoma is positively correlated with OSCC lymph node metastasis and clinical stage.The results suggest that Rb pathway exists in oral cancer and is closely related to the prognosis of oral cancer.It is one of the important indicators to evaluate the prognosis of oral cancer.(2)The results of this study show that E2F1 is highly expressed in oral squamous cell carcinoma,but there is no significant correlation with clinical pathological parameters.The expression of E2F1 and RbAP48 in oral cavity There is no significant correlation between squamous cell carcinoma,and the exact molecular mechanism of action is still unclear,and further research and discussion are needed.
