Objective To detect the expression of 14-3-3?in oral squamous cell carcinoma and analyze its association with clinicopathological parameters.Also,to detect the expression of the important factors of the PI3K/AKT pathway Akt and the pro-apoptotic factor Bad of the Bcl-2 family and explore their clinical significance.Methods The expressions of 14-3-3?in 86 cases of OSCC and Akt and Bad in 15cases of OSCC were detected by immunohistochemistry.The correlation between the expression of 14-3-3?and clinical pathological parameters was analyzed by chi-square test(?~2test).Results(1)The results of immunohistochemical examination of 86 cases of oral squamous cell carcinoma showed that 14-3-3?was mainly expressed in the cytoplasm of OSCC tissue,and yellow or brown granules are visible in the cytoplasm.Its positive expression rate was 51.2%(44/86).The positive expression rate of 14-3-3?was 50.0%(30/60)in the male group and 53.9%(14/26)in the female group.There was no significant difference between the two groups(P>0.05).The positive expression rate of 14-3-3?in the age group?60 years was 54.9%(28/51),and in the age group<60 years was 45.7%(16/35).There was no significant difference between the two groups(P>0.05).In the T classification,the positive expression rate of 14-3-3?in the T1 group was 39.1%(9/23),59.5%(25/42)in the T2 group,50.0%(6/12)in the T3 group and 44.4%(4/9)in the T4 group.There were no statistically significant difference among the groups(P>0.05).In tumor differentiation,the positive expression rate of 14-3-3?was 65.2%(15/23)in the well-differentiated group,40.0%(18/45)in the high-medium differentiated group,60.0%(9/15)in the moderately differentiated group and 66.7%(2/3)in the poorly differentiated group.There were no significant difference among the groups(P>0.05).The positive expression rate of 14-3-3?in the lymph node metastasis positive group was 48.2%(26/54),and the lymph node metastasis negative group was 56.3%(18/32).There was no statistically significant difference between the groups(P>0.05).However,in the clinical stage,the positive expression rate of 14-3-3?in the stage?group was 22.2%(2/9),the stage?group was 75.0%(12/16),and the stage?group was 63.0%(17/27).In the IV group,it was 38.2%(13/34).There were statistically significant differences among the clinical staging groups(P<0.05).The results suggest that the expression of 14-3-3?in OSCC has a negative correlation with clinical stage,but has no significant correlation with the gender and age of the patients,the T classification,the lymph node metastasis,and the degree of tumor differentiation.(2)The results of immunohistochemical examination of 15 cases of oral squamous cell carcinoma showed that the Akt protein was mainly expressed in the cytoplasm and nucleus of OSCC tissue,with a diffuse distribution of yellow or brown-yellow granules.Besides,the cell nucleus was deeply stained with brownish yellow.Its positive expression rate was 40.0%(6/15).The Bad protein was mainly expressed in the cytoplasm of OSCC tissues,with a focal distribution of yellow or brown-yellow.Its positive expression rate of 26.7%(4/15).Conclusions(1)The expression of 14-3-3?in oral squamous cell carcinoma has a negative correlation with clinical stage.The results suggest that 14-3-3?has a certain role in the progress of OSCC;(2)The important factor of PI3K/AKT pathway Akt and Bcl-2 family member Bad are expressed in oral squamous cell carcinoma.From this result,it is speculated that the mechanism of 14-3-3?in OSCC may be related to the apoptosis effect of the Bcl-2family and the PI3K/AKT pathway,which needs to be further explored.
