Objective:The aim of the study was to detect the differences in expression of mRNA and protein of?1 adrennergic receptor(?1-AR)and?2 adrennergic receptor(?2-AR)in oral squamous cell carcinoma(OSCC)tissues,the corresponding para-carcinoma and normal oral mucosa tissues,and to investigate the clinical significance.Methods:60 cases of OSCC containing 30 cases of pathological diagnosed lymphatic matastasis and 30 cases of pathological diagnosed non-lymphatic matastasis were included in this research,and the clinicopathological and prognostic data of the patients were collected;15 cases of normal oral mucosa andcorrespondingpara-carcinomatissuesweretakenascontrol.Hematoxylin-eosin(HE)staining was performed on the samples of carcinoma tissues and para-carcinoma tissues.RT-qPCR and Western blot were used respectively to detect the expression of mRNA and protein of?1-AR and?2-AR in tissues of OSCC,the corresponding para-carcinoma and those of normal oral mucosa;The method of 2~-??CTT??CTT was used to calculate the relative expression of mRNA of?1-AR and?2-AR;The gray value of protein bands was measured by Image Lab Software to represent the expression of protein of?1-AR and?2-AR in tissues of OSCC,the corresponding para-carcinoma and those of normal oral mucosa;the differences in expression of mRNA and protein of?1-AR and?2-AR in OSCC tissues,the corresponding para-carcinoma and normal oral mucosa tissues were analyzed by SPSS softwares'Independent-Samples t Test and paired-samples t test;The relationships among the differences in expression of mRNA and protein of?1-AR and?2-AR in OSCC tissues and the clinicopathological and prognostic data were investigated by the method of Independent-Samples t Test.Results:HE staining results showed that there were obvious keratin pearls in the nests of carcinoma tissues and no carcinoma cells were observed in para-carcinoma tissues.RT-qPCR results showed that(1)the relative expression of mRNA of?1-AR and?2-AR in OSCC tissues was obviously higher than that in normal oral mucosa tissues;(2)there was no significant difference between the relative expression of mRNA of?1-AR in OSCC tissues and that in the corresponding para-carcinoma tissues.The relative expression of mRNA of?2-AR in OSCC tissues was obviously higher than that in the corresponding para-carcinoma tissues.Western blot results showed that(1)the expression of protein of?1-AR and?2-AR in OSCC tissues was obviously higher than that in normal oral mucosa tissues;(2)the expression of protein of?1-AR in OSCC tissues was higher than that in the corresponding para-carcinoma tissues.The expression of protein of?2-AR in OSCC tissues was obviously higher than that in the corresponding para-carcinoma tissues.Correlation analysis of?1-AR and?2-AR and the clinicopathological and prognostic data showed that(1)the expression of mRNA and protein?1-AR was correlated with the clinical stage whereas it had no significant correlation with gender,age,tumor size,tumor pathological grade,lymph node metastasis,and postoperative recurrence;(2)the expression of mRNA and protein?2-AR was correlated with the clinical stage,lymph node metastasis,postoperative recurrence whereas it had no significant correlation with gender,age,tumor size and tumor pathological grade.Conclusions:1.The expression of mRNA and protein of?1-AR and?2-AR in OSCC tissues was higher than that in normal oral mucosa tissues.2.There was no significant difference in the expression of?1-AR mRNA between OSCC tissues and corresponding para-carcinoma tissues,but the expression of?1-AR protein in OSCC tissues was higher than that in corresponding para-carcinoma tissues;The expression of mRNA and protein of?2-AR in OSCC tissues was obviously higher than that in the corresponding para-carcinoma tissues.3.The expression of?1-AR was correlated with the clinical stage whereas it had no significant correlation with gender,age,tumor size,tumor pathological grade,lymph node metastasis,and postoperative recurrence.4.The expression of?2-AR had no significant correlation with gender,age,tumor size and tumor pathological grade whereas had sigificant correlated with the clinical stage,lymph node metastasis,postoperative recurrence.Patients with higher expression of?2-AR tend to own higher possiblity of recurrence and lymph node metastasis...
