| Objective:In this study,fluorescence quantitative PCR was used to observe and compare the application of self-made traditional Chinese medicine Shengyufang retention enema to treat Bifidobacterium,Enterococcus,and fragile in the intestinal flora of patients with mild to moderate ulcerative colitis(UC).The changes in the number of Bacteroides were explored to explore the mechanism of action of Shengyufang in the treatment of ulcerative colitis,which provided a theoretical basis for the clinical promotion of Shengyufang.Methods: 30 patients with mild to moderate UC who were admitted to the Affiliated Hospital of Shandong University of Traditional Chinese Medicine from January 2018 to December 2018,and 30 healthy volunteers were selected and divided into experimental group and control group.Depending on the extent of the lesion,the experimental components were the rectal group(rectal UC patients)and the non-rectal group(left colonic and total colonic UC patients).The rectal group was given Shengyufang retention enema treatment;the non-rectal group was given Shengyufang retention enema combined with mesalazine enteric-coated tablets for oral treatment.The stool samples of the experimental group were collected before and 14 days after the administration,and only the stool specimens were collected once in the control group.The Mayo scores before and after treatment were recorded in the control group,and the bacterial DNA in samples was tested by FQ-PCR,the content of Bifidobacterium,Enterococcus and Bacteroides fragilis in the sample was obtained.Compare and analyze the Mayo score and FQ-PCR results of theexperimental group,the rectal group and the non-rectal group.Results:(1)The experimental group,rectal group and non-rectal group were compared with the Mayo score after treatment,The mean value of the Mayo score before treatment was 6.50 points in the experimental group and 2.67 points after 14 days of treatment.The mean value of the Mayo score before treatment was 6.83 points in the rectal group and 3.00 points after 14 days in the treatment group.The mean value of the Mayo score before treatment in the non-rectal group was 6.42 points,2.58 points after 14 days of treatment,the scores after treatment were significantly lower than those before treatment(P<0.05).Compared with the control group,the number of Bifidobacterium and Bacteroides fragilis in the stool samples before treatment in the experimental group,the rectal group and the non-rectal group was significantly decreased(P<0.05);the number of Enterococcus was significantly increased(P<0.05).).Compared with pre-treatment,the number of Bifidobacteria and Bacteroides fragilis in the stool samples of the experimental group,the rectal group and the non-rectal group was significantly higher than that before treatment(P<0.05);Enterococcus and pre-treatment The specific number was significantly reduced(P<0.05).(2)There was no significant difference in the Mayo score and the sample FQ-PCR test between the rectal group and the non-rectal group(P>0.05).It was proved that oral mesalazine enteric-coated tablets had no significant effect on this test.Conclusion:Traditional Chinese medicine Shengyufang retained enema to treat UC significantly;Shengyufang enema treatment of UC may be through regulating intestinal flora. |
