Objectives To study the effect of methadone on intestinal microflora in mice,the structural and quantitative changes of intestinal microflora in mice after gastric perfusion with methadone were analyzed,as well as the law of change and mechanism of action.It is expected to provide experimental reference for patients with methadone maintenance therapy in intestinal microflora changes,and to provide reference for the diagnosis and treatment of intestinal diseases in patients with methadone maintenance therapy.Methods After 3 days of adaptation,75 mice were randomly divided into 3 groups with 25 mice in each group.and then 3 groups were administered methadone,oat ?-glucan and normal saline,0.3m L/d,and mice are free to eat and drink water.1.During gastric perfusion,the hair,diet,defecation,mental state and activity of the mice were observed,the weight of the mice was measured every 7 days.2.After 28 days of gavage,the mice were killed and the feces of the colon and rectum in the mice were collected,then we extract bacterial genome DNA.The total flora,Bacteroides,Bifidobacterium,Lactobacillus,Escherichia coli and Enterococcus in the intestinal tract of mice were quantitatively detected by real-time fluorescence quantitative PCR.3.Collecting feces from the colon and rectum of mice,The traditional culture method was used to cultivate the feces,anaerobic bacteria and fungi,and the colonies were counted.4.Statistical analysis: the SPSS16.0 software was used to analyze whether there were statistical differences in the body weight,the number of bacteria and the number of colonies among the three groups.Results1.During the period of feeding and gavage,the mice generally grew well,with bright and smooth hair,normal diet and urine,good activity,no depression or excessive exuberance,and no death during feeding.There was no significant difference in survival status among the three groups.Over time,the weight of the three groups kept increasing steadily.The body weight of mice in different periods had significant difference(P<0.05),while there was no significant difference in body weight among different groups(P>0.05).2.There was significant difference in the number of aerobic and anaerobic bacteria among the three groups(P<0.05).Among them,oat ?-glucan group had the highest number of colonies,followed by methadone group,and normal saline group was the least.There was no significant difference in the number of fungal colonies among the three groups(P>0.05).There was significant difference in the number of aerobic bacteria and anaerobes in the group compared with that in normal saline group(P<0.05),and the number of aerobic bacteria and anaerobic bacteria in methadone group was less than that in normal saline group.The number of aerobic and anaerobic colonies in oat ?-glucan group and normal saline group were significantly different(P<0.05),and the number of aerobic and anaerobic colonies in oat ?-glucan group was higher than that in normal saline group.3.The difference of total intestinal flora,Bacteroides and Escherichia coli between methadone and normal saline group was statistically significant(P<0.05),and thetotal intestinal flora,Bacteroides and Escherichia coli in methadone group were more than those in normal saline group.The number of total intestinal flora,Bacteroides,Lactobacillus and Escherichia coli in oat ?-glucan group and normal saline group was significantly different(P<0.05),and oat ?-glucan Group of total intestinal flora,Bacteroides,Lactobacillus,Escherichia coli more than normal saline group.4.There was no significant difference in the ratio(B/E value)among the three groups of Bifidobacteria and Escherichia coli(P>0.05).However,the value of B/E was less than 1,indicating that the resistance of the mice in each group decreased.Conclusions1.Methadone and oat ?-glucan had no effect on body weight,diet and activity in mice.2.The number of aerobic bacteria in methadone group was less than that in normal saline group,and the number of aerobic bacteria and anaerobic colony in oat ?-glucan group was more than that in normal saline group.3.The number of total intestinal flora,Bacteroides and Escherichia coli in methadone group was more than that in normal saline group,and the total intestinal flora,Bacteroides,Lactobacillus and Escherichia coli in oat ?-glucan group was more than those in normal saline group.4.The intestinal colonization resistance of the three groups of mice decreased.
