Objective:To investigate the expression and significance of serum NO and NOS positive neurons in myenteric plexus in experimental terminal ileitis.Methods:Sixty male Sprague-Dawley rats were picked and randomly divided into 3 groups,which were recorded as control group,suture group and model group.Model group:after anesthesia,the side-to-side anastomosis of the terminal ileum and the cecum was adopted to the ileum-colonic reflux model.Suture group:after anaesthesia,surgical incisions and sutures were made in the same placeas as the model group.Control group:after anesthesia,the rats were expected to wake up normally.At the the 2nd week and 8th week after operation,half rats of each group were randomly selected.Collecting the serum,then we used chemical method to measure the serum NO concentration.Collecting the terminal ileitis tissue,we observed the tissue with naked eyes first,then histopathologic changes of half of tissue were observed after hematoxylin-eosin staining.The other half of tissue use Immunohistochemistry to detect the expression of NOS positive neurons.The changes and correlations of the above indicators were analyzed.Results:1.Visual and microscopic observation:after 2nd weeks,the inflammation scores in the model group and the suture group were higher than those in the control group?P<0.05?.after 8th week,there was no noticeable difference in scores between the suture group and the control group?P>0.05?.The model group had higher inflammation scores than the control group and the suture group.?P<0.05?.The scores in the model group were higher in the 8th week than in the 2nd week,and it in the suture group were lower in the 8th week than in the 2nd week,the difference was statistically significant?P<0.05?.2.Serum NO concentration:after 2nd week,Serum NO concentrations in the model group and the suture group were higher than those in the control group?P<0.05?.after 8th week,there was no noticeable difference between the suture group and the control group?P>0.05?.The serum NO concentration in the model group was higher than that in the control group and the suture group?P<0.05?.The concentration in the model group were higher in the8th week than in the 2nd week,and it in the suture group were lower in the 8th week than in the 2nd week?P<0.05?.3.The expression score of NOS positive neurons in the myenteric plexus:after 2nd weeks,the expression scores of NOS positive neurons in the model group and the suture group were higher than those in the control group?P<0.05?.After 8th weeks,there was no noticeable difference in scores between the suture group and the control group?P>0.05?.The expression scores of the model group were higher than that of the control group and the suture group?P<0.05?.The scores in the model group were higher in the 8th week than in the 2nd week.And the scores in the suture group were lower in the 8th week than in the 2nd week,the difference was statistically significant?P<0.05?.4.Correlation analysis:The serum NO concentration and NOS positive neuron expression's scores in the myenteric plexus are positively associated with the degree of inflammation?r1=0.90,r2=0.78,P<0.01?.The serum NO concentration are positively associated with NOS positive neuron expression's scores.Conclusion:Theconcentration of serum NO and theexpression of NOS positive neurons in the myenteric plexus are increased in ETI,they have a positive correlation with the degree of inflammation of terminal ileitis.Both NO and NOS positive neurons in the myenteric plexushave important significance in the pathogenesis of ETI.
