Background:Pelvic inflammatory disease(PID)is a common cause of infertility,chronic pain and ectopic pregnancy as the inflammation and infection of female upper genital tract.Among them,the incidence of sexually transmitted diseases related to pelvic inflammatory disease has gradually increased in recent years.Ozone water,also known as O3,was proved as a clearly effective mean for treating inflammation.In this paper,we constructed a PID rat model to explore the efficacy of O3 pelvic injection in the treatment of PID.Methods: To investigate the effect of O3 on PID,a rat model of acute PID was established.50 SD rats were randomly divided into 5 groups,10 in each group,namely control group,saline group(NS group)and levofloxacin group(LVLX group).45 ug/mL O3 group and 60 ug/mL O3 group.The control group did not do any treatment.The NS group received pelvic injection of 20 ml/kg/d NS,the LVLX group received pelvic injection of 0.06 g/kg/d LVLX,and the remaining two groups of O3 groups received pelvic injection at 45 ug/mL and 60 ug/ mL of O3.Drug injection was continued for 7 days,during which 3 rats were randomly selected for ultrasound examination.After 7 days,the blood was taken,and the uterus was taken out from the rats.The damage and repair of the uterus were observed under light microscope.The levels of IL-2,IL-6 and TNG-? inflammatory factors in the serum of each group were detected by ELISA.The immunological indicators were tested by single radial diffusion(SRD)for IgG,IgA,IgM,C3 and C4,and acid-?-naphthyl acetate esterase(ANAE)for T lymphocyte transformation and rosette formation.Immunohistochemis-try was used to detect differences in IL-6 expression in various uterine tissues.Results:1.Ultrasonography can also be used to aid in diagnosis of PID and direct PID treatment.Before modeling,the pelvic cavity of rats had no obvious abnormal lesion.After successful modeling,the lump boundary and capsule in PID rats were obscure,and weak echo or strong echo could be observed in the capsule.The dense light spots,the obvious dark areas of the liquid.Compared with the control group,there was no significant change in the ultrasound imaging of the NS group after treatment,but the pelvic cavity was significantly improved in the LVLX group,the 45 ug/mL O3 group,and the 60 ug/mL O3 group.2.The levels of inflammatory factors in serum were detected by ELISA.Compared with the control group,IL-2,IL-6 and TNF-? in NS group did not change significantly(p>0.05,n=10),while group LVLX and group 45 ug/mL O3 and In group 60 ug/mL O3,IL-2 increased,IL-6 and TNF-? levels decreased(p>0.05,n=10),and the difference was statistically significant.3.SRD and ANAE were used to detect the immunological parameters in the serum of rats.Compared with the control group,the immunoglobulin content and immune function of the rats in the NS group did not change significantly(p>0.05,n=10).The contents of IgG,IgA,IgM,C3 and C4,T lymphocyte transformation rate and rosette formation rate were significantly increased in the LVLX group,45ug/mL O3 group and 60ug/mL O3 group(p>0.05,n=10),and the difference was statistically significant..4.HE staining was used to observe endometrial damage and repair.In the control group and NS group,scattered or stratified inflammatory cell infiltration and epithelial cell degeneration and necrosis were observed.Compared with the control group,the endometrial inflammation of the LVLX group,45ug/mL O3 group and 60ug/mL O3 group was significantly improved,the boundary between the mucosa and the myometrium was clear,the epithelial structure was basically integrated,and there was a large amount of inflammatory cell infiltration in the myometrium.5.Immunohistochemistry was used to detect the expression of IL-6 in uterine tissue.Compared with the control group,there was no significant change in NS group(p>0.05,n=10),while IL-6 expression in LVLX group and O3 group decreased(p<0.05,n=10).Conclusion: Studies have shown that O3 can inhibit the inflammatory response of PID rats,improve the immunoglobulin content and immune function of PID rats,thereby improving PID rats and providing a new solution for the treatment of PID.
