Abrus cantoniensis Hance,a kind of plant in Abrus Adans,Leguminous,also known as Guangdong Acacia.It is a local crude medicine in the Guangdong and Guangxi regions.It is also a kind of traditional Chinese medicine collected by 2015 edition of Chinese Pharmacopoeia.The medicinal part is the dried whole herb after remoivng pods.Abrus cantoniensis Hance tastes bitter,cool,has the effect of eliminating dampness and yellowing,clearing away heat and detoxifying,soothing liver and stopping the pain.Studies have shown that Abrus cantoniensis Hance is rich in chemical components and has a wide range of pharmacological effects.It is mainly used for the treatment of hepatobiliary diseases in clinic.It is a good medicine for treating acute hepatitis and is commonly used in cooking soup in folk.However,in the last 10 years,few studies have been carried out on the chemical constituents of Abrus cantoniensis Hance.Existing studies have shown that the active ingredients are mainly terpenoids and flavonoids.Up to now,more compounds isolated from the whole grass are triterpenoids,but flavonoids are less studied,even there is no report on isoflavones.In view of this,this thesis systematically studies the chemical constituents of Abrus cantoniensis Hance by useing modern chromatographic andspectroscopy techniques to in order to obtain some novel structural compounds,further enrich the chemical composition diversity of Abrus Adans,and provide a basis for later generations to study the material basis of its efficacy.In this paper,using silica gel column chromatography,thin layer chromatography,reversed phase ODS column chromatography,Sephadex LH-20 column chromatography,semi-preparative reversed phase HPLC and other chromatographic methods,14 monomers were isolated from70% ethanol extract of Abrus cantoniensis Hance.The structure of the 11 compounds were identified by UV,IR,1D/2D NMR,HR-ESI-MS.They are butesuperin A-7''-O-?-glucopyranoside(1*),8-O-methylretusin-7-O-?-D-apifuranosyl-(1?2)-?-D-glucopyranoside(2*),8-methylretusi-7-O-?-D-glucopyranoside(3),4',7-di-O-methylpuerarin(4),7,4'-dihydroxy-8-methoxyisoflavone(5),7-hydroxy-8,4'-dimethoxy isoflavone(6),8-hydroxy-7,4'-dimethoxy-isoflavone(7),3-hydroxy-9-methoxy pterocarpan(8),isobiflorin(9),7,4'-dihydroxy-8-methoxy isoflavone(10),protocatechuic acid ethyl ester(11).Among them,compounds 1* and 2* are new compounds,the former belonging to new isoflavonolignan compound,the number of which reported by SciFinder are only 7.Compound 4 is also the second found isoflavonelignan glycoside so far.Compounds 1 to 7 were all isoflavones,which were isolated from Abrus cantoniensis Hance for the first time;Compounds 3,7,and 8 were isolated from Abrus Adans for the first time.In this paper,isoflavones were isolated from Abrus cantoniensis Hance for the first time,including 2 new compounds,especially compound 1* was a novel isoflavonelignan compound,which provided an experimental basis for isolating isoflavones from Abrus cantoniensis Hance.It enriches the chemical composition diversity of Abrus Adans and Abrus cantoniensis Hance,and provides reference for the subsequent discovery of more novel isoflavones.
