Objective: The microvascular density and cell proliferation activity in common epithelial tumors of lacrimal gland were measured and counted by the method of immunohistochemical staining.The expression of Epidermal Growth Factor like domain7(EGFL7)protein in the common lacrimal gland epithelial tumors and normal lacrimal gland tissue was determined by immunohistochemical staining method at the same time.The correlation of EGFL7 with tumor microvascular density(MVD)and tumor cell proliferation activity in lacrimal gland epithelial tumors was discussed.Method: A total of 46 paraffin-embedded specimens of common lacrimal gland epithelial tumors treated with lacrimal gland tumors and 10 cases of normal lacrimal gland,were collected in the Third Clinical College of Qingdao Municipal Hospital Affiliated to Qingdao University Eye Center from January 2009 to January 2019,lacrimal gland tumors including 20 cases of lacrimal gland pleomorphic adenoma,12 cases of pleomorphic adenocarcinoma,and 14 cases of adenoid cystic carcinoma.HE staining was performed on all tissue samples,and the expression of EGFL7 protein in normal lacrimal gland tissue,pleomorphic adenoma,pleomorphic adenocarcinoma,and adenoid cystic carcinoma was compared by immunohistochemistry.At the same time,the tumor microvascular were labeled with CD34 antibody,and the tumor MVD in all specimens was calculated.The proliferative activity of tumor cells was represented by Ki67,and the correlation between the expression of EGFL7 protein and MVD and Ki67 in lacrimal epithelial tumors was analyzed.Results: EGFL7 protein is mainly expressed in the cytoplasm of tumor cells of lacrimal gland pleomorphic adenocarcinoma and adenoid cystic carcinoma,that is,the cytoplasm is brown or brown colored in positive cells.Immunohistochemical staining showed that EGFL7 was not positively expressed in normal lacrimal gland tissue.The positive expression rate of EGFL7 in lacrimal gland pleomorphic adenoma,in lacrimal gland pleomorphic adenocarcinoma,and in lacrimal adenoid cystic carcinoma were 5.0%(1/20),83.33%(10/12),and 85.71%(12/14),respectively.The expression rate in both malignant tumors was significantly higher than that in pleomorphic adenomas and normal lacrimalgland tissues(p<0.001).CD34 can stain tumor microvascular in a brown-yellow single or cluster cell populations.The expression of CD34 in lacrimal gland pleomorphic adenocarcinoma(32.58±14.46)and adenoid cystic carcinoma(43.43±4.60)was significantly higher than that of pleomorphic adenoma(4.20±1.19)(p<0.001).Ki67 is brownish in the cell nucleus with proliferative activity.The expression of Ki67 in lacrimal gland pleomorphic adenocarcinoma(44.83±13.68)and adenoid cystic carcinoma(26.29±8.44)was significantly higher than that of pleomorphic adenoma(2.80±3.14)and normal tissue(0.40±0.70)(p<0.001).And the expression of EGFL7 protein was positively correlated with MVD and Ki67 in lacrimal epithelial tumors(r=0.897,p<0.001;r=0.837,p<0.001).Conclusion: This study examined the expression of EGFL7 in common lacrimal epithelial tumors and analyzed the correlation between the expression level of EGFL7 in lacrimal epithelial tumors and tumor MVD and Ki67.Through experimental analysis,the following conclusions can be obtained:1.The expression of EGFL7 in both malignant tumors was significantly higher than that in pleomorphic adenomas and it is positively correlated with the expression of MVD,indicating that the high expression of EGFL7 plays an important role in promoting tumor angiogenesis.2.There was a significant positive correlation between the expression of EGFL7 and the expression of Ki67 in lacrimal epithelial tumors,suggesting that the up-regulation of EGFL7 was closely related to tumor proliferation.3.EGFL7 plays an important role in tumor angiogenesis and proliferation in malignant lacrimal epithelial tumors,providing a new potential target for the treatment of lacrimal epithelial tumors.
