Objective:1.The expression levels of the Second mitochondrial activating factor(Smac)and Cysteine aspartate specific protease-3(caspase-3)in endometrial adenocarcinoma,proliferative endometrium and atypical hyperplasia endometrium were detected at tissue level.2.The relationship between the expression of Smac and caspase-3 and the clinicopathological features of endometrial adenocarcinoma was analyzed.and the correlation between Smac and caspase-3 in endometrial adenocarcinoma.Methods:Immunohistochemical method SP was used to detect the expression and positive localization of Smac and caspase-3 in 95 cases of paraffin specimens(17 cases in the proliferative endometrium group,20 cases in the endometrial dysplasia group,and 58cases in the endometrial adenocarcinoma group).The experimental data were collected.The relationship between the expression of Smac and caspase-3 and the clinicopathological features in endometrial adenocarcinoma was analyzed by SPSS21.0 statistical software.The relationship between Smac and caspase-3 was analyzed by Spearman correlation analysis.The correlation between the two in endometrial adenocarcinoma was statistically significant at P<0.05.Results:1.The positive expression rate of Smac in the endometrial proliferative phase,endometrial atypical hyperplasia group and endometrial adenocarcinoma group decreased in turn,the difference was statistically significant(?~2=12.704,P<0.05).2.The positive expression rate of caspase-3 in the endometrial proliferative phase,endometrial atypical hyperplasia group and endometrial adenocarcinoma group decreased in turn,and the difference was statistically significant(?~2=10.522,P<0.05).3.The positive expression of Smac was associated with surgical pathological stage,different histological differentiation and lymph node metastasis(P<0.05),and was not related to patient age and myometrial invasion depth(P>0.05).4.The positive expression of caspase-3 was only related to histological differentiation(P<0.05),and was not associated with age,surgical pathological stage,depth of myometrial invasion and lymph node metastasis(P>0.05).5.There was a significant positive correlation between the expression of Smac and caspase-3 in endometrial adenocarcinoma(r=0.378,P<0.05).Conclusions:1.The positive expression rates of Smac and caspase-3 in the endometrial proliferative phase,endometrial atypical hyperplasia group and endometrial adenocarcinoma group were decreased,which may be related to the occurrence of endometrial adenocarcinoma.2.In the endometrial adenocarcinoma group,Smac showed less positive expression rate in patients with lower degree of tissue differentiation,higher surgical stage,and lymph node metastasis.The expression of caspase-3 was only related to the degree of tissue differentiation,and both As the degree of tissue differentiation decreased,the expression decreased,indicating that Smac and caspase-3 may be involved in the malignant development of endometrial adenocarcinoma.3.There was a positive correlation between the expression of Smac and caspase-3in endometrial adenocarcinoma,indicating that the combined detection of these two proteins has certain significance for the occurrence and development of endometrial adenocarcinoma.
