Traditional Chinese medicine is a valuable asset accumulated by the Chinese nation and disease for thousands of years.At present,the inheritance and development of traditional Chinese medicine faces problems such as unclear clinical positioning and unclear mechanism of action,which hinders the effective use and secondary development of drugs in clinical practice.Xefp granules are traditional Chinese medicine preparations for children with spleen and stomach qi deficiency,indigestion,and physical emaciation.The prescription is based on the classic prescription “Shenling Baizhu San” in "Prescriptions People's Welfare Pharmacy",which has the function of invigorating the spleen and stomach and helping digestion.The prescription is mainly composed of baizhu,chenpi,shanzha,dangshen,lianzi,fuling.Modern pharmacological studies have shown that Xefp granules has a regulating effect on gastrointestinal motility,yet its absorbable components,main acting tissues,potential new indications Problems such as the mechanism of action need to be resolved.In this paper,based on tissue localization,disease link location and comparison of similarity of listed drugs,the clinical positioning of Xefp granules was carried out.To clarify the modern scientific basis for its treatment of diseases,to propose its clinically appropriate conditions,and to provide reference for clinical use.Taking the Xefp granules as an example to study the method of clinical positioning of traditional Chinese medicine.Objective1.Predicting the absorbable components,mechanism and clinical indications of Xefp granules,to provide specific direction for clinical localization and innovative drug development of Xefp granules;2.Exploring new methods of network pharmacology research.Method1.Collecting the components of Xefp granules from TCMID and TCMSP databases.The components with good medicinal properties were screened by ADMET.Targets and scores of absorbable components were collected from BATMAN-TCM,PUBCHEM and STITCH databases.Integrating targets from three sources and evaluating the possibility of targets comprehensively;2.Using UP_TISSUE tool of DAVID database to locate the tissue of X Xefp granules;According to the disease link,explore the mechanism of Xefp granules on "same treatment for different diseases" for indigestion and depression;Based on the similarity of compounds at the target and pathway level,the clinical application and mechanism of Xefp granules were analyzed by comparing with the listed western medicines for treating indigestion and depression.3.Expanding the application of the above methods.Studying on the mechanism of Glxbbx decoction by tissue enrichment.Studying on the mechanism of Yueju Wan in the treatment of dyspepsia with depression by disease association.Result1.126 absorbable components of Xefp granules were initially obtained by ADMET screening;372 potential targets with high reliability were obtained by integrating BATMAN-TCM,PUBCHEM and STITCH databases.2.The enriched main functional tissues are in the brain and liver;The mechanism of Xefp granules for "same treatment for different diseases" of indigestion and depression may interact with neuroactive ligand-receptor interaction,response to organic substance,ion channel activity,homeostatic process,circulatory system process,and regulation of cAMP biosynthetic process,blood circulation,calcium signaling pathways,GABA receptor activity,dopamine binding,adrenoceptor activity,regulation of smooth muscle contraction and other biological processes;The composition of Xefp granules and the listed western medicines for clinical treatment of indigestion and depression are grouped into three categories: a.Antagonism of 5-HT3 receptor and activation of 5-HT4 receptor;b.Blocking the H2 receptor.c.Antagonism of 5-HT3 receptor.And all three drugs have antidepressant effects.In addition,Xefp granules may also be related to biological process such as vitamin A metabolic process,prostaglandin E receptor activity,icosanoid metabolic process,inflammatory response,purine metabolism,aldosterone-regulated sodium reabsorption,chronic myeloid leukemia,steroid hormone receptor activity,neuroprotection,regulation of monooxygenase activity,pathways in cancer.3.The method of tissue localization was applied to Glxbbx decoction,and its location was mainly in the heart and liver.The mechanism may be related to heart rate regulation,cardiomyocyte survival,immune inflammation,vascular smooth muscle contraction,lipid metabolism.The method of locating disease links is used in the study of Yueju Wan,and its treatment of indigestion with depression is mainly related to neuron apoptotic process,neurotrophin signaling pathway,serotonergic synapse,bile secretion,gastric acid secretion.ConclusionBased on the three methods of tissue orientation,disease link orientation and similarity comparison of listed drugs,the Xefp granules were analyzed from different angles on solving the problems of absorbable components,main functional tissues,potential new indications,and mechanism.To provide new ideas and methods for network prediction analysis of traditional Chinese medicine and ethnic medicine.
