Objective: The expression of Protein tyrosine phosphatase non-receptor type 12(PTPN12)and the mRNA relative expression of PTPN12 were detected in renal cell carcinoma tissues and corresponding normal tissues of patients with renal cell carcinoma(RCC).Meanwhile,the clinical data was analyzed,which laid a foundation for further study of the mechanism of PTPN12 gene in renal cell carcinoma.Methods:1.PV immunohistochemical method was applied to detect the expression of PTPN12 protein in renal cell carcinoma tissues and corresponding normal tissues of 58 patients with renal cell carcinoma(RCC).2.The RT-qPCR(real-time quantitative PCR)method was applied to detect the relative expression of PTPN12 mRNA in renal cell carcinoma and corresponding normal tissues of 58 patients with renal cell carcinoma.3.The clinical data Including age,gender,primary tumor size and clinical stage of 58 patients with renal cell carcinoma was recorded,and the relationship between the PTPN12 protein expression and the relative expression of PTPN12 gene mRNA and the clinical data was analyzed.Results:1.The positive expression rates of PTPN12 protein in RCC tissues and the corresponding normal tissues were 29.3% and 75.9% in 58 specimens of renal cell carcinoma.The positive expression rate of PTPN12 protein in RCC tissues was significantly lower than that in the corresponding normal tissues(P < 0.05).There was no significant correlation between the positive expression rate of PTPN12 protein in RCC tissues of 58 patients with RCC and clinical data including the age,gender,tumor diameter and clinical stage of the patients(P>0.05).2.The mRNA relative expression of PTPN12 in RCC tissues and the corresponding normal tissues of 58 patients with RCC were 0.60±0.89 and1.00±0.01.The mRNA relative expression of PTPN12 in RCC tissues was significantly lower than that in corresponding normal tissues(P <0.05).There was no significant correlation between the mRNA relative expression of PTPN12 in RCC tissues of 58 patients with renal cell carcinoma and clinical data including the age,gender,tumor diameter and clinical stage of the patients(P>0.05).Conclusion:1.The expression of PTPN12 protein in RCC tissues was significantly lower than that of normal tissues,suggesting that the decrease of expression of PTPN12 protein may be related to the occurrence of RCC.There was no obvious correlation between the positive expression rate of PTPN12 protein in RCC tissues and the clinical data including age,gender,tumor diameter and clinical stage of the patients.2.The mRNA relative expression of PTPN12 in RCC tissues is significantly lower than that of the corresponding normal tissues,suggesting that the decrease of mRNA relative expression of PTPN12 may be related to the occurrence of RCC.There was no obvious correlation between the mRNA relative expression of PTPN12 in RCC tissues and the clinical data including age,gender,tumor diameter and clinical stage of the patients.
