| Objective: To investigate the expression of phosphatidylinositol 3-kinase p85 a subunit(PI3Kp85a),EZRIN and tumor suppressor gene chromatin remodeling complex SWI/SNF subunit(AT rich interactive domain 1A: ARID1A)in thyroid cancer and adjacent normal tissues,and to explore the relationship among PI3Kp85 a,EZRIN and ARID1 A.The relationship between PI3Kp85 a,EZRIN and ARID1 A expression and pathological factors was discussed in order to clarify the significance of PI3Kp85 a,EZRIN and ARID1 A expression in thyroid carcinogenesis,invasion and metastasis.Methods: From January 2017 to May 2018,114 cases of thyroid cancer which include 51 males and 63 females aged 25-61 years(39.6 ±11.0 years old)and 43 cases of normal thyroid tissue which include 17 males and 26 females,aged 28-65 years(42.7 ±12.0 years old)were collected from the patients who were treated in the Department of mammary gland and thyroid surgery of Zaozhuang Municipal Hospital.The expressions of PI3Kp85 a,EZRIN and ARID1 A in 69 cases of papillary thyroid carcinoma,18 cases of follicular carcinoma,15 cases of undifferentiated carcinoma,12 cases of medullary carcinoma and 12 cases of normal thyroid tissues adjacent to thyroid carcinoma were detected by immunohistochemistry,and PI3Kp85 a,EZRIN and ARID1 A were detected by RT-PCR.The expression level of RNA(SPSS19.0 software was used for statistical analysis);the expression of PI3Kp85 a,EZRIN and ARID1 A in different thyroid tissues was analyzed by t test and chi-square test;the correlation between the expression of ARID1 A and EZRIN,PI3Kp85 a was analyzed by Spearman grade correlation test.Results: 1.Compared with the expression of PI3Kp85 a,EZRIN ARID1 A mRNA and Protein positive in normal adjacent tissues,the expression of PI3Kp85 a and EZRIN in thyroid cancer tissue increased significantly(P < 0.01),while the expression of mRNA and protein positive expression rate of ARID1 A in thyroid cancer tissue decreased significantly(P < 0.01).2.The expression of ARID1 A mRNA was the highest in papillary thyroid carcinoma(0.67±0.39),which was significantly higher than that in poorly differentiated follicular carcinoma(0.50±0.33),undifferentiated carcinoma(0.38±0.29)and cephaloma(0.52±0.54)(P < 0.05).The higher the malignancy,the lower the expression of ARID1 A mRNA,and the negative correlation was found between the expression of ARID1 A mRNA and the degree of malignancy(P < 0.05 r =-0.689).3.The expression of PI3Kp85 a and EZRIN mRNA in papillary thyroid carcinoma was significantly increased and positively correlated with the malignant degree of thyroid carcinoma.(P <0.05 r=0.625)4.Positive expression of PI3Kp85 a,EZRIN and ARID1 A in thyroid carcinoma of different pathological classification: papillary carcinoma:63.8%,53.7%,55.1%;follicular carcinoma:72.2%,66.7%,44.4%;undifferentiated,thyroid carcinoma:86.7%,86.6%,33.3%;medullary thyroid carcinoma: 75.0%,75.0%,41.7%.Conclusion: PI3Kp85 a,EZRIN and ARID1 A play an important role in the occurrence,invasion and metastasis of thyroid carcinoma.The high expression of mRNA and its protein of PI3Kp85 α and EZRIN protein in thyroid carcinoma tissue and its low expression in normal thyroid tissue suggest that the high expression of both may be associated with the pathogenesis of thyroid cancer,and provides a solid theoretical basis to the invasion and transfer of thyroid cancer.The deletion or decrease of ARID1 A can promote the high expression of PI3Kp85 alpha and EZRIN protein,and then promote the occurrence,development and invasion of thyroid carcinoma.The prognosis of invasion and metastasis can provide a basis for the evaluation and treatment of thyroid cancer. |
