Objective: To study the relationship between the expression of Arid1 a protein and the pathogenesis of gastric cancer and its pathological features.Methods: Selected the patients with gastric cancer underwent curative surgical resection with standard lymphadenectomy and partial gastrectomy for benign lesions from 2012 to 2015 at Wannan Medical College First Affiliated Yijishan Hospital in China.And excluded mucosal cancers(T stage 1a)and multiple simultaneous gastric cancers.Gastric cancer specimens(included paracancerous tissue)and normal gastric mucosa tissue fragments were immunohistochemical staining.The primary antibodies was rabbit monoclonal anti-ARID1A(Clone number:EPR13501).Under the guidance of the two pathologists to determine the results of each section.Patients' electronic medical records were reviewed for clinicopathologic information such as gender,age,tumor location,TMN stage,tumor depth,lymph node metastasis,tumor differentiation,presence of vascular and nerve invasion,and Lauren classifications.The pathologic stages were adjusted based on the American Joint Committee on Cancer Staging Manual,7th edition.The results were analyzed using statistical methods.Results: A total of 42 cases of normal gastric mucosal,and 104 cases of gastric cancer tissue immunohistochemical staining were observed in this study;include 75 cases of paracancerous tissue.The loss of Arid1 a exprssion in normal gastric mucosa,paracancerous tissue and gastric carcinoma was 4.76%(2/42);8.00%(6/75)and 23.08%(24/104),respectively.The loss of Arid1 a exprssion was highest in gastric cancer tissue.The differences was statistically significant(normal gastric mucosal vs gastric cancer tissue,P=0.008;paracancerous tissue vs gastric cancer tissue,P=0.008).There was no significant correlation between the loss of Arid1 a exprssion and gender,age,smoking and alcohol consumption(P(29)0.05).The deletion of Arid1 a was associated with previous history of gastric ulcer,and the difference was statistically significant(P=0.048).By analyzing the results of immunohistochemical staining of gastric cancer,there was no correlation between the loss of Arid1 a exprssion and the location of tumor,the classification of Lauren,the tumor differentiation and the presence of nerve invasion(P(29)0.05).In the depth of tumor infiltration,the experimental results show that T3+T4 gastric cancer cases accounted for 63.46% of the proportion.The deeper the tumor infiltration,the higher the negative rate of Arid1 a protein expression.The negative expression rate of Arid1 a protein was 30.00% and 33.33% in T3 and T4 stage gastric cancer patients,respectively,which was significantly higher than that in T1 stage(6.67%)and T2 stage(8.70%),the difference was statistically significant(P <0.05).The more the lymph node metastasis,indicate that the higher loss of Arid1 a expression in gastric cancer.The negative expression rates of Arid1 a protein in N2 and N3 gastric cancer tissues were 31.82% and 41.38% respectively,which were significantly higher than those of N0 and N1.The loss of Arid1 a protein expression in gastric cancer patients with vascular invasion was higher than that without it,and the difference was statistically significant(P <0.05).The negative rate of Arid1 a protein expression in patients with tumor diameter bigger than 5cm was 42.42%,higher than the tumor less than or equal to 3cm(5.88%)and greater than 3 less than or equal to 5cm(21.62%)of patients with gastric cancer.The difference was statistically significant(P <0.05).Conclusion: The rate of loss of Arid1 a protein expression is higher in gastric cancer patients with previous history of gastric ulcer,suggesting that the expression of Arid1 a protein was associated with gastric ulcer.The loss of Arid1 a expression was associated with tumor size,depth of invasion,lymph node metastasis and vascular invasion.It suggested that loss of Arid1 a protein expression in gastric cancer patients has a poor biological behavior of the tumor.The loss of Arid1 a expression may be related to the occurrence of gastric cancer,suggesting that the Arid1 a gene may be a tumor suppressor gene.
