Objective:To investigate the characteristics of EB virus infection in children with chronic tonsillitis and adenoid hypertrophy,and to explore the status of EB virus infection and the relationship between EB virus latent infection and chronic tonsillitis and adenoid hypertrophy in children,and the mechanism that may cause related tumors,and Provide basic data for future EB virus related research.Methods:The pathological specimens of 40 patients with chronic tonsillitis who underwent surgical removal of tonsil adenoids and 50 specimens of adenoids in children with adenoidal hypertrophy were collected.Immunohistochemistry and in situ hybridization were used to detect paraffin tissue of surgically resected specimens.The expression of Epstein-Barr virus latent membrane protein 1(EBV-LMP1)and Epstein-Barr virus encoding RNA(EBER)in the sections was observed and the cell proliferation was observed under a light microscope.The peripheral blood T lymphocyte subsets in children with chronic tonsillitis were detected By flow cytometry.The changes of peripheral blood T lymphocyte subsets in children with chronic tonsillitis in EB virus positive and negative children were compared.Results:1.The positive rate of LMP1 in children with chronic tonsillitis was 45%,and the positive rate of LMP1 in adenoid hypertrophy group was 66%.The positive expression of LMP1 was statistically significant in the two groups(P<0.05).2.The positive rate of EBER in children with chronic tonsillitis was 47.5%,the positive rate of EBER in adenoid hypertrophy group was 70%,and the positive rate of EBER was statistically significant(P<0.05).3.Immunohistochemistry and in situ hybridization were used to detect the positive rate of EBV,There is no difference between the two,P>0.05.4.The positive rates of LMP1 and EBER were not statistically significant in latent infections of different genders,ages and EBV.5.Under light microscope,the surface of the LMP1-positive tonsil tissue was covered with stratified squamous epithelial cells.The lymphoid tissue in the lamina propria was proliferated.The EBV-positive cells were dispersed in the follicular and germinal centers,and the positive signal was localized in the cytoplasm.EBER-positive tonsil and adenoid tissue and structure are intact,and the EBER positive signal is localized in the nucleus of the cell.6.90 cases of chronic tonsillitis and adenoid hypertrophy were detected by immunohistochemistry and in situ hybridization.Single positive expression included LMP1+EBER-and EBER+LMP1-,the expression rate was 18.97%,double positive The expression rate of LMP1+EBER+ accounted for 81.03%(P<0.05).7.There was no significant difference in the ratio of CD3+T,CD4+T,CD8+ T lymphocytes and CD4+/CD8+ T cells between the EBV positive group and the control group(P>0.05).Conclusions:1.Children with chronic tonsillitis,adenoid hypertrophy EBV infection rate is high,the nasopharynx is an important latent part of EBV.Epstein-Barr virus LMP1 and Epstein-Barr virus-encoded small RNA(EBER)can be present in non-tumor tissues.2.The latent infection rate of EBV in children with adenoid hypertrophy is higher than that in children with chronic tonsillitis.3.Epstein-Barr virus LMP1 and Epstein-Barr virus-encoded small RNA(EBER)can be present in non-tumor tissues.4.Children with chronic tonsillitis and adenoid hypertrophy are mostly latent type Ⅱ and type Ⅲ,and a few are latent 0 and type Ⅰ.
