Objective:To check the serum monocyte chemotactic protein 1(MCP-1),interleukin-17A(IL-17A)and interleukin-10(IL-10)by cytometric bead arry(CBA),and analysis the correlation between serum MCP-1,IL-17 A,IL-10 and the degree of NAFLD fatty liver to provide direction for early screening of NAFLD.Methods:A total of 75 NAFLD patients and 15 healthy persons were included in the study,and the 75 NAFLD patients were divided into 21 with mild NAFLD,27 with moderate NAFLD and 27 with severe NAFLD.We measured the height,weight,waist circumference and hip circumference of subjects,then calculated body mass index(BMI)and waist-hip ratio(WHR).We also collected fasting elbow venous blood and then serum hepatic enzymes,lipids,blood sugar and insulin were detected by laboratory,We calculated HOMA-IR according to those data and apply CBA to detect the serum MCP-1,IL-10 and IL-17 A.Using SPSS20.0 statistical software to process the above data.Results:(1)baseline information was compared in NAFLD and NC group:the BMI,WHR,HOMA-IR in NAFLD group were higher than NC group and the serum ALT,AST,?-GT,CHE,TG,LDL-C,FPG,MCP-1 and IL-17 A levels showed the same trend meanwhile the serum HDL-C and IL-10 levels were lower than NC group,all these showed(P<0.05).the other indicators include gender distribution,age,the serum ALP,TC andFINS levels had no significant difference(P>0.05).(2)baseline information was compared in different NAFLD grades and NC group:the degree of fatty liver in NAFLD group was related to BMI,WHR,HOMA-IR,the serum ALT,AST,?-GT,CHE,TG,LDL-C,HDL-C and FPG levels.BMI,HOMA-IR,the serum ALT,AST,ALP,TG,LDL-C,HDL-C and FPG levels in different degrees of NAFLD group had significant differences(P<0.05)compared with NC group,which could be used to differentiate non-fatty liver,mild,moderate and severe fatty liver group.(3)serum cytokine levels was compared in different NAFLD grades and NC group:the degree of fatty liver in NAFLD group was related to serum levels of MCP-1,IL-17 A and IL-10.Compared with healthy control group,there were significant differences in serum MCP-1,IL-17 A and other indicators between different degrees of NAFLD group(P<0.05),which could be used to identify normal liver,mild,moderate and severe NAFLD group.the serum MCP-1 level in severe NAFLD group was higher than that in mild and moderate NAFLD group and healthy control group(P<0.05),but there was no significant difference between mild and moderate NAFLD group(P>0.05).the serum IL-17 A level in severe NAFLD group was higher than that in mild NAFLD group and healthy control group(P<0.05),but there was no significant difference between mild and moderate NAFLD group(P>0.05).the serum IL-10 level in severe NAFLD group was lower than that in healthy control group(P<0.05),but there was no significant difference between NAFLD groups(P>0.05).(4)the correlation of MCP-1,IL-17 A and IL-10 with baseline data: the serum MCP-1 level was positively correlated with BMI,HOMA-IR,serum ALT,AST,?-GT,TG and LDL-C levels(P<0.05).The serum IL-17 A level was positively correlated with BMI,WHR,HOMA-IR,serum ALT and FINS levels(P<0.05),mennwhile was negatively correlated with serum HDL-C levels(P<0.05).The serum IL-10 level was negatively correlated with BMI,HOMA-IR,serum CHE and FINS levels(P<0.05).(5)HOMA-IR,MCP-1,IL-17 A and IL-10 were positively correlated with the severity of fatty liver,while serum IL-10 was negatively correlated with the severity of fatty liver(P<0.05).Conclusion:Serum levels of MCP-1,IL-17 A and IL-10 are correlated with the degree of fatty liver in patients with NAFLD,which is a possible direction for non-invasive diagnosis of NAFLD.
