Objective:To determine DNA ploidy analysis,HR-by measuring the detection rate of DNA ploidy,high-risk human papilloma virus(HR-HPV)and routine pathological examination of pelvic lymph nodes in cervical squamous cell carcinoma The relationship between HPV detection and routine pathological examination in different clinicopathological groups of cervical squamous cell carcinoma,and the relationship between DNA ploidy and HR-HPV and lymph node micrometastasis of cervical squamous cell carcinoma,and the clinical significance of individualized squamous cell carcinoma of the cervix Treatment and individualized follow-up provide guidance.Method:1.From November 2017 to November 2018,27 patients with cervical squamous cell carcinoma underwent surgical treatment in gynecology department of the First Hospital of Shanxi Medical University.513 fresh pelvic lymph nodes were collected.The lymph nodes were divided into lymph node metastasis group(experimental group1)and lymph node non-metastasis group(experimental group 2)according to the results of routine pathological examination by hematoxylin and Eosin(HE)staining.2.DNA ploidy analysis was performed on all lymph nodes of experimental group 1 and 2 by DNA-AICM detection system.3.HR-HPV was detected in all lymph nodes of experimental group 1 and 2 by conduction hybridization.4.Statistical analysis was performed with SPSS25.0 system analysis software.Two tests were used for comparison among groups.Result:1.The positive rates of DNA ploidy,HR-HPV and DNA ploidy combined with HR-HPV were compared among 513 lymph nodes detected.There were significant differences between the staging groups of cervical squamous cell carcinoma(P < 0.01)(Table 1).With the increase of clinical stages,the positive rates of DNA ploidy,HR-HPV detection and DNA ploidy combined with HR-HPV detection increased.2.Among 513 lymph nodes detected,there was no significant difference in the positive rates of DNA ploidy,HR-HPV and DNA ploidy combined with HR-HPV among age groups of cervical squamous cell carcinoma(P > 0.05)(Table 1).3.In the experimental group 1,107 lymph nodes were detected in 5 patients.In the experimental group 2,there were 22 patients,406 lymph nodes were detected.The positive rate of DNA ploidy and HR-HPV in group 1 was higher than that in group 2,and the difference was statistically significant(P < 0.01)(Table 2).4.The positive rate of DNA ploidy and HR-HPV in the positive lymph nodes of the experimental group 1 was significantly higher than that in the negative lymph nodes of the experimental group 1(P < 0.01)(Table 2).5.The positive rates of DNA ploidy and HR-HPV in the negative lymph nodes of the experimental group 1 were higher than those in all the lymph nodes of the experimental group 2(P < 0.01)(Table 2).Conclusion:1.DNA aneuploidy and HR-HPV have different positive expression rates in negative lymph nodes by routine pathological examination,suggesting that micrometastasis may exist in negative lymph nodes by routine pathological examination.2.The positive rates of DNA ploidy and HR-HPV were different in different stages of cervical squamous cell carcinoma lymph nodes.The higher the clinical stage of cervical squamous cell carcinoma,the higher the positive detection rate of DNA aneuploidy and HR-HPV.3.The positive rates of DNA ploidy and HR-HPV in cervical squamous cell carcinoma lymph nodes were different among different age groups.The results showed that the positive rates of DNA aneuploidy and HR-HPV were independent ofage.4.The positive rate of DNA ploidy and HR-HPV in group 1 was higher than that in group 2,which indicated that the higher the positive rate of DNA ploidy and HR-HPV in patients with lymph node metastasis by routine pathological examination.5.The positive detection rate of DNA ploidy and HR-HPV in the positive lymph nodes of group 1 was higher than that of the negative lymph nodes of group 1.It indicated that DNA aneuploidy and high expression of HR-HPV were more likely to occur in the metastatic positive lymph nodes of group 1 by routine pathological examination.6.The positive detection rate of DNA ploidy and HR-HPV in the negative lymph nodes of group 1 was significantly higher than that of all the lymph nodes in group 2,indicating that DNA aneuploidy and HR-HPV were more easily detected in the metastasis group than in the non-metastasis group.It further suggests that lymph node micrometastasis may occur earlier in metastatic group.
