Objective:Melaleuca alternifolia,also known as the qianchenta or Australian Tea Tree belonging to the Melaleue of Myrtaceaefamily,Is Natived toparts of New Wales and New Zealand of Australia,Contained inthe European Pharmacopeia,Australian native plants and The Illustrated Herb Encyclopedia,Melaleuca alternifolia is mainlyused to treat skin inflammationbefore.Later itwas found has strong anti-bacterial and anti-inflammatory physiological activities.TeaTree Oil is active ingredient of the Melaleuca alternifolia.The latest studies showed that tea tree oilof Melaleuca alterniflorawas importantrole related to anti-in fl ammatory,andantibacterial activities.Tea Tree Oil,light yellow to colorless oily liquid.,isobtained by steam distillation.Based on the above content,theaimofthistopic wasto optimize theMelaleuca alternifolia provenances and growth environment,to explore the extraction methods andthecomponents contentofTea Tree Oil,as well as todevelop a stable temperature-sensitive Gynecological gel,.This projectprovide ideas for the comprehensive application of Melaleuca alternifolia.Method:Using GC method,the effects of different water content,different provenances,different planting environments and different time factors were analyzed by using the essential oil yield and the relative content of each component as the index.Taking the yield of tea tree oil and the relative content of each component as the evaluation index,the single factor method and the orthogonal experiment method are preferred to extract the essential oil by steam distillation,supercritical extraction and solvent method;the single factor method is preferred for the pilot testofProduction Process.The star point design method is used to optimize the preparation process of bitter tea gynecological gel.The membrane release model was used to evaluate the in vitro release mode of the gel,the rheological study was used to evaluate the fluid properties of the gel,the HPLC method was established to determine the effective components of the gel,and the MIC and MBC concentrations of the gel were determined by the two-fold dilution method.The double-dilution method was used to determine the MIC concentration of 10 kinds of pathogenic bacteria by extracting the aromatic water and the dregs aqueous extract.Result:The yield of fresh products of Tea tree oil was 8.11% higher than dry products,and there was no significant difference in quality.According to the quality standard of tea tree oil,the pros and cons of the provenance: Nandu provenance > Linkeyuan provenance > cotton Xinkan provenance > lotus pond provenance,field plant > mountain plant;evaluation of provenance byessential oil yield: Lin Keyuan Source > Liantang provenance > Nandu provenance > Rongshan provenance > cotton Xinkan provenance,field plant > mountain plant.Accordingthecomprehensive evaluation of the provenance,Nandu is thebest planted in the field.The relative content of terpinol-4-alcohol reached the maximum in May,and the relative content curve showed a conical distribution.The relative content of 1-8 eucalyptus decreased month by month after reaching the maximum value in January.The tea tree essential oil prepared by steam distillation is in line with the international tea tree essential oil standard,and the tea tree essential oil prepared by the supercritical extraction method and the solvent method has a large difference with the international tea tree essential oil standard.The in vitro membrane release model showed a zero-order release of the gel in vitro;rheological studies showed that the gel met the requirements of vaginal administration;HPLC effectively controlled the quality of the gel;MIC and MBC concentrations showed that the gel had strong antibacterial activity.Sterilization effect.Aromatic water has an inhibitory effect on some oral pathogenic bacteria;the water extract solution of Melaleuca ternata has a better inhibitory effect on most pathogenic bacteria.Conclusion:The cultivation of the Melaleucaalternifolia can be carried out by the source of Nandu.The standardized farmland planting mode and the supply of organic fertilizer contribute to the formation of essential oils.According to the process research,the key factor affecting the yield of essential oil is steam temperature(steam pressure),which is the heat energy required for the essential oil to volatilize.The higher the temperature,the faster the efficiency;steam distillation,supercritical extraction,solvent preparation The tea treeoilthatmade by thethreedifferent methodshas a large difference in composition and cannot be substituted for each other.The prepared gel has strong antibacterial effect and stable quality.The oil extraction by-product aromatic water and dregs aqueous extract still have a strong inhibitory effect on most bacteria,and provide basic data for the comprehensive utilization development of Melaleuca alternifolia.
