Wuji Baifeng cream according to Wuji Baifeng capsule formula,composed of twelve traditional Chinese medicinal materials,Wuji,Dihuang,dandan,baishao,huangqi nourish blood,nourishing and other functions.The original quality standard of Wufeng ointment was as follows: Radix Salviae miltiorrhiza,Radix Ginseng,Glycyrrhiza uralensis,determination of Paeoniflorin content.With the development of the cause of traditional Chinese medicine,in order to achieve the requirements of stable product quality,safety and effectiveness,and strong compliance.The subject of this paper is to improve the quality standard of black chicken white Phoenix ointment.On the basis of the original standard identification of three kinds of medicinal materials,the identification of Radix Rehmanniae,astragalus membranaceus and radix astragali was adde.Determination of Danshensu Sodium and Paeoniflorin in the same chromatographic conditions.Qualitative identification of characteristic map.A method for the determination of oligopeptide in Wuji Baifeng ointment was established.The quality standard of Wuji Baifeng has been drawn up.The main research contents are as follows:(1)The TLC identification of Radix Rehmanniae,Radix Astragali and Rhizoma Cyperi was added in this project,and the identification method of ginseng in the original standard was optimized.Under the same TLC condition,ginseng and Radix Astragali could be identified simultaneously.The TLC identification method of six herbs(Radix Rehmanniae,Radix Astragali,Rhizoma Cyperi,Radix Salviae Miltiorrhizae,Radix Ginseng and Radix Glycyrrhizae)in the prescription was completed,without negative interference,and had good stability It conforms to the relevant provisions of the analysis method of the quality standard of Chinese Pharmacopoeia.(2)The content of Danshensu and paeoniflorin was determined simultaneously under the same liquid phase condition.The preparation methods of the samples were investigated.The chromatographic conditions including detection wavelength,stationary phase,mobile phase composition and column temperature were investigated.The final determination conditions were verified by methodological investigation and three batches of samples.The established method for the determination of Danshensu and paeoniflorin was simple and feasible.It conforms to the relevant provisions of the analysis method of the quality standard of Chinese Pharmacopoeia.(3)In this paper,protein was converted into oligopeptides by enzymatic hydrolysis of black chicken.According to the national standard-GB/T22729-2008 oligopeptide determination,the determination of protein content in Wuji Baifeng ointment minus.Tagpre-column derivatization method.The extraction solvent of free amino acid from the sample,the extraction method and the selection of purification agent were investigated,and the chromatographic conditions were optimized.Finally,the method for the determination of oligopeptide in Wuji Baifeng ointment was studied.The results showed that the results of each test in the methodology were good.The oligopeptide content of three batches of Wuji Baifeng ointment was determined.The oligopeptide content determination method could be used for the quality control of Wuji Baifeng ointment.(4)Study on the characteristic atlas of Wuji Baifeng ointment by HPLC,The characteristic chromatogram of Wuji Baifeng ointment was established.18 characteristic peaks were identified from five kinds of medicinal materials: Radix Ginseng,Radix Paeoniae Alba,Radix Bupleuri,and six peaks of Salvianolic acid B,Paeoniflorin and glycyrrhizin were identified,which provided experimental basis for product quality and quality control.
