The statutory quality standard of Xieriga-4 is contained in the PharmaceuticalStandards of the Ministry of Health of the People's Republic of China?Mongolian Pharmaceutical Branch?.This prescription is composed of turmeric,phellodendron,gardenia and Tribulus terrestris.It has anti-inflammatory,analgesic and antipyretic effects.For urinary arrest,lower urinary tract infection,bladder prickling,our group studied the network pharmacology of Xieriga-4 earlier,established the network model of chemical composition-target-disease of Xieriga-4,and preliminarily clarified the interaction and binding mode between Xieriga-4 and prostatic disease-related targets through molecular docking theory,and established the anti-inflammatory active ingredients of Xieriga-4.In order to further explore the anti-inflammatory effect of Xieriga-4,a rat model of non-bacterial prostatitis was established.The pharmacokinetics of the anti-inflammatory active ingredients in the serum of normal rats and non-bacterial prostatitis model rats were studied by serum pharmacochemistry.The metabolism of total terpenoids and alkaloids in normal rats and non-bacterial prostatitis model rats was studied.This paper is divided into four parts to study the pharmacodynamic material basis of Mongolian medicine compound Xieriga-4.The first part Separation and enrichment of the main chemical constituents of Mongolian medicine Xieriga-4.Enrichment of macroporous adsorbent resin the effective fraction of Xieriga-4 was separated,and the chemical constituents consistent with its main function were further analyzed and identified.The substances obtained in the earlier stage of the research group were enriched,which laid a foundation for the basic research of the effective substances of Xieriga-4.Identification by physicochemical identification and modern spectroscopic analysis techniques(MS,1H-NMR,13C-NMR,etc.)structure of the compound.Six compounds were isolated and enriched from Mongolian medicine Xieriga-4.They were alkaloids:berberine,phenolic acid compounds:curcumin,noroxycurcumin,dimethoxycurcumin,terpenoids:gardenoside,citrin compounds:Phellodendron.The second part studies the anti-inflammatory effect of Xieriga-4.Through the network pharmacology and molecular docking research,therelationship between the anti-inflammatory active ingredients and the target of inflammation disease was preliminarily clarified.Now this paper further confirms the anti-inflammatory effect of Xieriga-4 by discussing the therapeutic eect of Xieriga-4 on non-bacterial prostatitis.The results showed that Xieriga-4,a Mongolian medicine compound,had a therapeutic effect on non-bacterial prostatitis in rats induced by complete Freund's adjuvant.Compared with model control group,Xieriga-4 had an inhibitory effect on non-bacterial prostatitis?P<0.05?,and Xieriga-4 had the best effect.The serum levels of TNF-a,PSA,IL-4,testosterone?T?,estradiol?E2?and prostaglandin E2?PGE-2?in tissue abrasive fluid were measured by ELISA.The results showed that the levels of TNF-a,PSA,IL-4,E2 and PGE-2 in Xiega-4 groups decreased and increased.Compared with the model control group,there were significant differences in the high and medium dose groups of Xieriga-4.The difference was significant?P<0.01?.Paraffin section was used for pathological examination of Xieriga-4 experimental animals.The pharmacological effects of Xieriga-4,a Mongolian medicine compound,were further explored by pathological examination results.The results showed that the inflammatory characteristics of the model group were alleviated in the high,middle and low dose group of Xieriga-4.The effect of the medium dose group was better,and the number of inflammatory cells was significantly reduced,which was similar to that of the positive control group.In the third part Study on the in vivo constituents of Compound Xieriga-4Terpenoids,alkaloids,limonoids,phenolic acids and volatile oils are the mainchemical constituents in the active fraction of Xieriga-4.A lot of pharmacological studies on terpenoids,alkaloids,phenolic acids,limonoids and their monomers have been carried out at home and abroad.The results show that the above components have anti-inflammatory effects and are the material basis of anti-inflammatory,analgesic,clearing heat and eliminating stasis of Xieriga-4.The content determination and pharmacokinetics of phenolic acid anti-inflammatory active ingredients in serum of normal rats and non-bacterial prostatitis rats were studied earlier in our group.A method for determination of total terpenoids and alkaloids anti-inflammatory active ingredients in serum of normal rats and non-bacterial prostatitis rats was established by modern means.Pharmacokinetics of total terpenoids and alkaloids from Xieriga-4 in serum of normal and non-bacterial prostatitis rats.A study on the metabolism of total terpenoids and alkaloids in normal rats and non-bacterial prostatitis model rats was established.The therapeutic effect of Mongolian medicine Xieriga-4 on non-bacterial prostatitis was clarified,and the scientific basis for the pharmacodynamic substance of Xieriga-4was provided.
