OBJECTIVE: To study the expression specificity of Hsp70 and hMAM in different grades of breast cancer patients,and to analyze the relationship between the expression of these two proteins in breast cancer tissues and the corresponding clinicopathological features,and provide new ideas for clinical research on breast cancer immunotherapy.Exploring the role of Hsp70 and hMAM in breast cancer provides a basic theory for early diagnosis and prognosis of breast cancer patients.Methods: 1.Patients with breast cancer were excluded from previous radiotherapy,chemotherapy and neoadjuvant treatment,and the interference of drugs and other factors on hMAM and Hsp70 in breast cancer was excluded.Clinical patient information was collected from 90 patients with breast cancer,including age,menstrual status,tumor size,clinical stage,histological grade,and lymph node metastasis.2.90 cases of breast cancer tissue paraffin-embedded blocks(60 cases of breast cancer tissues and 30 cases of adjacent breast tissues)were arranged.The pathological sections were used to detect the expression of Hsp70 and hMAM in tumor tissues by immunohistochemical staining.3.SPSS19.0 statistical software was used to analyze the relationship between the specificity of Hsp70 and hMAM expression in breast cancer tissues and the age,menstrual status,tumor size,clinical stage,histological grade,and lymph node metastasis of breast cancer patients.RESULTS: The positive expression of Hsp70 and hMAM detected by immunohistochemistry showed that the positive rate of hMAM was 71.7% in breast cancer and the positive rate of Hsp70 was 65%.The positive rate of hMAM in normal breast tissue was 3.3%,and the positive rate of Hsp70 was 13.3%,the difference was statistically significant(P<0.05).Both Hsp70 and hMAM showed an increasing trend in the development of normal breast tissue to breast cancer.The expression of hMAM and Hsp70 and various clinicopathological factors showed that the expression level of hMAM was not correlated with age,menstrual status and tumor size(P>0.05),which was related to lymph node metastasis,clinical stage and histological grade(P<0.05).The expression level of Hsp70 wasnot correlated with age,menstrual status and histological grade(P>0.05),but was related to tumor size,clinical stage and lymph node metastasis(P<0.05).And the higher the clinical stage,the higher the positive expression of Hsp70.When the expression of hMAM and Hsp70 in breast cancer tissues was analyzed by Spearman correlation,the correlation coefficient was r=0.547,P<0.05,and it was considered that there was a positive correlation between the positive expression of hMAM and Hsp70.Conclusion: 1.Hsp70 and hMAM are highly expressed in breast cancer,but are not expressed in normal breast tissue,and because hMAM is expressed almost exclusively in breast cancer,and has high specificity and sensitivity,it can be used for early diagnosis.Judging prognosis provides an important reference value.2.Analysis of Hsp70 and hMAM expression and multiple clinicopathological factors showed that hMAM expression level was not related to age,menstrual status,tumor size,but related to lymph node metastasis,clinical stage and histological grade;Hsp70 expression level and age The menstrual status and histological grade were not related,but related to tumor size,clinical stage,lymph node metastasis,and the higher the clinical stage,the higher the positive expression of Hsp70.Both expressions are inseparable from the infiltration of breast cancer metastasis and can be used for clinical diagnosis.3.In this study,the expression of Hsp70 and hMAM is positively correlated,indicating that they are involved in the metastasis and infiltration of breast cancer,then the combined detection of Hsp70 and hMAM expression in breast cancer tissue can guide the comprehensive treatment of breast cancer.It is helpful in the diagnosis and identification of metastatic malignant tumors of unknown origin.
