Objective 1) To study the significance of the expression of hMAM protein in breast cancer initially; 2)To investigate the expression of hMAM, HER-2/neu,p53 and nm23 in breast cancer and their correlation with each other; 3) To explore the relationship of hMAM with biological characteristics of breast cancer preliminarily.Materials and methods 103 patients with breast cancer undergone treatment at Xiangya Hospital of Central South University between February 2004 and September 2006 were chosen randomly into experiment group,while 10 breast fibroma tissues and 28 normal breast tissue beside the focus of carcinoma were chosen into contrast group respectively. Those breast cancer patients have no history of operation or benign leision relating with breast and were not given any chemotherapy or radiological therapy. The surgical or BAND NEEDLE biopsy specimens underwent immunohistochemical analysis for hMAM. The patient's clinicopathological characteristics were obtained through collecting medical records. Statistical analyses were performed using SPSS software version 13.0. Analysis of single factors was assessed with x~2 testing or t testing. Non-parametric correlations were established using Spearman rank correlation testing. The Logistic Regression Model was used to assess the correlation of hMAM protein with HER-2/neu, nm23, p53 and clinicopathologic characteristics .Result Of the 103 patients,86(83.4%) showed the positive expression of hMAM in breast cancer tissue, while no hMAM expression in 28 normal breast tissue and only one breast fibroma has positive hMAM expression , of which the difference was statistical significantly. 84.6%(72/81) breast cancer patient of early stage were positive in the expression of hMAM ,which was higher than that of later stage(63.6%). In the age group younger than 35,80%(8/10) patients were positive in the expression of hMAM , while there were 93.0% and 69.4% respectively in the age groups ranging from 36 to 50 and elder than 50 .All the difference above has statistical significance (p<0.05) . The expression of hMAM in breast cancer tissue was not significantly related to other cliniclpathological characteristics, including BMI,the frequency of pregnant and labor,the state of menopause,the size of tumor,the pathological type of tumor , the status of axillery lymph node metastasis, the nuclear grade, and the status of hermone receptor. As to the correlation between hMAM and HER-2/neu ,when "row×column"x~2 testing and non-parametric correlations analysis was implemented, the values of p are all less than 0.05.But there is no significant correlation between hMAM and nm23 as well as p53. In the Logistic Regression Model , the state of HER-2/neu, the age range are included,x~2=8.015, p=0.018.Therefore,this formulated multiple linear regression formula has statistical significance.Conclusions 1) The expression of hMAM protein in breast cancer is fairly high and has fairly high specificity; 2)There is significant correlation between the expression of hMAM protein and Her-2/neu; 3)The expression of hMAM correlates with different clinical staging and age range; 4) The expression of hMAM protein in breast cancer did not appear to correlate with clinicopathological characteristics of patients such as BMI,family history,pathological type,tumor size,the state of axillery lymph nodes ,the state of hermone receptor.
