| OBJECTIVE: Quality Standard of piperine,and gallic acid in the active ingredients of Mongolian medicine ultrafine powder of Nutmeg-5 Pills and conventional powder.Identification of common vladimiria root and inulae radix in the ultrafine powder of Nutmeg-5 Pill and conventional powder by Thin Layer Chromatography,and to provide scientific expenmental basis for the next step in quality standards.The dissolution amount of the ultrafine powder of Nutmeg-5 Pills and the active ingredients in the conventional powder were compared under the same conditions.METHODS: TLC was used to qualitatively identify vladimiria root,inulae radix,and piperine.The content of piperine and gallic acid was determined by high performance liquid chromatography(HPLC),and the dissolution amount was compared.The HPLC conditions for determination of piperine were as follows: column Waters C18(4.6 mm×250 mm,5μm),mobile phase: Methanol(A)-Water(B)=70:30,detection wavelength: 343 nm,column temperature: 25℃,flow rate: 1m L/min,injection volume: 10μL.The HPLC conditions for determination of gallic acid were as follows: column Waters C18(4.6 mmx250 mm,5μm),mobile phase: Methanol(A)-1% Phosphoric acid solution(B)=5:95,detection wavelength: 270 nm,column temperature: 25℃,flow rate: 1m L/min,injection volume: 10μL.RESULTS: After identification by thin-layer chromatography,the vladimiria root and inulae radix reference medicine and the piperine reference solution showed the same spots as the ultrafine powder and the conventional powder,respectively,and the negative control had no interference.The HPLC method was applied to the determination of the active constituents of the ultrafine powder of Nutmeg-5 Pill and conventional powder,the results showed that the piperine and gallic acid were in the range of 0.2137-1.1805μg and 0.1752~2.551μg, showing a good linear relationship.The average recovery rates were 95.38% and 97.84%.By comparing the dissolution amount of the ultrafine powder of Nutmeg-5 Pill and conventional powder,the results were as follows: the amount of the piperine and gallic acid in the ultrafine powder of Nutmeg-5 Pill pulp reached the highest value at 90 min and 120 min.They were respectively 1.1286 times and 1.1372 times of the conventional powder. |
