Objective: The expressions of Dicer and VEGF in villi tissues of embryo damage and normal villi were detected,and the differences and correlation between Dicer and VEGF expressions in the two groups were analyzed,and the correlation between Dicer and VEGF expressions and early embryo abortion were preliminarily analyzed,providing a new idea for discussing the pathogenesis of embryo abortion.Methods: A case-control retrospective study was used to select early pregnant womenin Qingdao municipal hospital from December 2017 to September 2018.20 patients with early embryo insemination were in the case group.And 20 cases of normal early pregnancy requiring induced abortion were randomly selected as the control group.Object inclusion criteria:(1)ultrasound shows a single intrauterine pregnancy sac and the women are the first pregnant with natural conception;(2)Both parents exclude chromosomal diseases;Clinical exclusion genital organic lesions;internal and surgical diseases,including hypertension,diabetes,heart disease,nervous system diseases;(3)Exclude TORCH infection,mycoplasma,chlamydia infection and acute or chronic inflammation of the reproductive system.(4)No endocrine abnormalities(5)Non-immune system disease;(6)No long-term oral medication;(7)the size of embryo or gestational sac was monitored by B-modeultrasound: Pregnancy(day)= embryo length(mm)+ 42(from7weeks to 12 weeks);Pregnancy(day)= mean internal diameter of pregnancy capsule(mm)+ 30(within 7 weeks),The actual gestational weeks were 7 to 10 weeks;(8)All patients included in the study had informed consent.All patients in the included group underwent artificial abortion,and the villi were preserved.Using the methods of western blotting and RT-PCR,the difference in expression level of Dicer and VEGF between the two groups were detected.All relevant data were analyzed using SPSS23.0.Result:(1)According to RT-PCR,the Dicer mRNA expression of the case group and the control group were 0.76±0.34 and 1.18±0.23,the VEGF mRNA expression of the case group and the control group were 0.83±0.57 and 1.99±0.83.The mRNA expression of Dicer and VEGF in the case group was lower than the control group,the difference was significant(p<0.05).(2)In western blotting,the Dicer expression of the case group and the control group were 0.17±0.01 and 0.37±0.02,the VEGF expression of the case group and the control group were 0.37±0.07 and 0.66±0.03.The expression of Dicer and VEGF in the case group was lower than the control group,the difference was significant(p<0.05).(3)The mRNA and protein relative expression of Dicer and VEGF were positive correlation(r=0.52?0.65;p<0.05).Conclusion:1.Decreased expression of Dicer in early villus is associated with embryonicdamage;2.Decreased expression of VEGF in early villus is associated with embryonic damage;3.Dicer in villi may play an important role in regulating vascular proliferation in villi during early pregnancy.
