Objective: B7-H3(CD276)is a type I transmembrane protein of the B7 family that is involved in the co-stimulatory and co-suppressive functions that regulate T cell responses.So far,a variety of tumor B7-H3 positive expression has been found,and B7-H3 levels are associated with tumor growth,invasion,metastasis,malignant stage and recurrence rate.In this paper,the expression of B7-H3 in prostate cancer tissues,combined with clinical medical records,to explore the relationship between B7-H3 and the development of prostate cancer,and to provide a better screening target and clinical targeted therapy.Theoretical basis.Methods: Real-time quantitative PCR(RT-PCR)and immunohistochemical techniques were used to detect the expression of B7-H3 mRNA and protein in48 cases of prostate cancer and 48 cases of benign prostatic hyperplasia,and the expression of B7-H3 was analyzed by combining clinical records.There was a statistical correlation between clinical stage,preoperative blood PSA level,gleason score,and cumulative survival rate after radical resection and tumor-free survival rate.results:1.RT-PCR results: The expression of B7-H3 in prostate cancer was significantly higher than that in benign prostatic hyperplasia(P<0.05).2.Immunohistochemistry results: Immunohistochemistry showed that B7-H3 protein was usually expressed on the cytoplasm or membrane,and the method of multiplying the degree of staining with the percentage of positive cells was evaluated.The pathologists with rich experience in reading were randomly selected from 10 high-power fields,and the positive staining of cell membrane or cytoplasm was positive.Among the 48 cases of cancer tissues,75.0%(36/48)were high expression,while 48 cases of benign prostatic hyperplasia were low expression,and the difference was statisticallysignificant(P<0.05).2.1.The protein expression of B7-H3 protein in prostate cancer was significantly higher than that in benign prostatic hyperplasia,which was consistent with the conclusion of RT-PCR.2.2.B7-H3 protein prostate cancer expression level was associated with clinical stage,preoperative blood PSA level,gleason score(P<0.05).2.3.By observing the immunohistochemical expression of prostate cancer specimens,combined with follow-up data analysis,it was found that there was no significant correlation between B7-H3 expression and postoperative cumulative survival rate in prostate cancer(P>0.05).There was a correlation between recurrence and survival(P<0.05).Conclusions:1.The mRNA and immunohistochemical expression of B7-H3 in prostate cancer tissues were higher than those in benign prostatic hyperplasia tissues.2.The expression of B7-H3 was correlated with clinical stage,preoperative PSA level and Gleason score of prostate cancer.3.There was no significant correlation between the expression of B7-H3 in prostate cancer and the cumulative survival rate.There was a correlation between the expression of B7-H3 in prostate cancer and the biochemical recurrence rate of tumor,and it was statistically significant;B7-H3 was high.Patients with lower expression of patients with prostatic cancer after radical resection are more likely to have biochemical recurrence and have a shorter biochemical recurrence time.
