Background and Objective: MYB proto-oncogene like 2(MYBL2)gene has been shown to be associated with poor prognosis in lots of tumors. However,it is unclear that the clinical significance of MYBL2 in prostate cancer(PCa).This study aimed to investigate the association of MYBL2 with tumorigenesis and prognosis in patients with PCa patients.Material and Methods: MYBL2 protein expression pattern in malignant phenotypes of human PCa and non-cancerous prostate tissue was demonstrated by immunohistochemical analysis.Meanwhile, the expression was validated by microarray-based Taylor and TCGA dataset at m RNA level. Then, the association of MYBL2 expression with the clinicopathological characteristics of patients with PCa was then statistically analyzed. Results: MYBL2 high expression was observed in clinical prostate cancer specimens related to non-cancerous prostate tissue according to the analyses of immunohistochemistry(IRS,PCa: 2.43±1.61 vs benign: 4.22±2.02,P=0.001).Linked the MYBL2 protein levels to the clinicopathological features of the patients, which showed the high expression of MYBL2 was significantly associated with high Gleason score(P=0.001).Also,Taylor and TCGA datasets have shown that the upregulation of MYBL2 m RNA expression in the PCa tissues significantly correlated to Gleason score, the presence of tumor metastasis, shorter overall survival and prostate-specific antigen(PSA)failure. What's more,as showed by univariate analyses, the upregulation of MYBL2 was a possible predictor or risk of poor biochemical recurrence(BCR)-free survival. Conclusions: Our findings disclose that MYBL2 may be an important role in the progression of PCa.More importantly, the increased expression of MYBL2 may potentially predict poor BCR-free survival in patients with PCa.
