Purposes:To explore the prognosis factors of locally advanced prostate cancer.To explore prognostic relevant abnormal gene expression and its underline mechanism.Substances and methods:Univariate and multivariate Cox regression analysis the value of factors in predicting the probability of biochemical recurrence.Through searching the public databases,looking for prognostic relevant abnormal gene expression.And confirm the result by immunohistochemical staining in our prostate tumor samples.Overexpression and Knockdown the target gene in prostate cancer cell lines to observe the changes of cell proliferation and migration.Explore the mechanisms by western blot and RT-PCR.Results: In multivariate analysis,specimen Gleason score and Early postoperative PSA retained independent significance(p<0.05).The nomogram predicting the risk of BCR showed relative good concordance index and good calibration.HOXD13 expression level is related to biochemical recurrence.HOXD13 expression level is an independent prognostic factor of biochemical recurrence.Low expression of HOXD13 promote prostate cancer metastasis via regulating BMP-SMAD pathway.Conclusions: The nomogram to predict the probability of BCR in patients with locally adverse pathologic features after radical prostatectomy showed good clinical applicability.This statistical tool may facilitate discussions at the point of treatment decision making.HOXD13 could be used as an independent prognosis factor in predicting biochemical recurrence after radical prostatectomy.HOXD13 suppresses prostate cancer metastasis via regulating SMAD1 transcription may contribute to disease progression.
