Objective To explore the role and the internal mechanism of SOX1 in cholangiocarcinoma,and offer a new ideas for the diagnosis and treatment of cholangiocarcinoma.Methods We performed Western blot,PCR and immunohistochemical to detect the expression of SOX1 in cholangiocarcinoma.We increase the expression of SOX1 by allowing cells to infect lentiviruses.The tumor invasion,metastasis and proliferation of cholangiocarcinoma cells were detected by Transwell,scratch test and CCK-8,plate clone assay.Double luciferase reporter gene,western blot,plasmid construction and other techniques were used to study the mechanism of SOX1 in cholangiocarcinoma.MSP,BSP technique was used to detect the methylation of promoter region of SOX1.Results We found that the expression of SOX1 was low in cholangiocarcinoma,and overexpression of SOX1 could inhibit the proliferation and invasion of cholangiocarcinoma,but had no effect on migration.Then we found that SOX1 can inhibit the phosphorylation of ERK to exert its anti-tumor effect.SOX1 also be regulated by mi R-155-5p,and mi R-155-5p can reduce the expression of SOX1.In cholangiocarcinoma,methylation occurs in the promoter region of SOX1,while in normal tissues,the methylation rate of the promoter region of SOX1 is lower,so the expression of SOX1 may be lower in cholangiocarcinoma due to the methylation of its promoter.Conclusion SOX1 can inhibit the cholangiocarcinoma through ERK pathway,SOX1 is regulated by mi R-155-5p,and SOX1 is hypermethylated in cholangiocarcinoma.Our research provides new ideas for the treatment of cholangiocarcinoma.
