| Objective:To measure the methylation level of sex determing region Y-box1(SOX1)in different cervical lessions,comparing the difference in the positive rate of methylation in each group of cervical lessions and analyzing its clinical signifincance.Methods:1.The cervical cancer tissues and matched adjacent tissues from 10 patients with cervical cancer were collected for quantitative analysis of SOX1 methylation level by Sequenom Mass ARRAY and Analyzing the difference between cervical cancer and the paired adjacent cancer.2.Expand the sample size to collect 386 cases of cervical lesion tissues and 554 cases of cervical lesion exfoliated cells,using QPCR method to qualitatively detect SOX1 gene methylation expression,and analyze the differences of each group of cervical lesions.Results:1.Sequenom Mass ARRAY quantitative detection of cervical cancer tissue SOX1 gene methylation sites the expression level of SOX1_Cp G_10,SOX1_Cp G_11,SOX1_Cp G_23,SOX1_Cp G_24,SOX1_Cp G_25.26.27.28.29.30,SOX1_Cp G_34.35,SOX1_Cp G_34.35,SOX1_Cp G_45.46 was significantly higher than paired cancer tissues,and the difference was statistically significant(P<0.05)2.In cervical lesions,the positive rates of SOX1 gene methylation in the cervical cancer group,high-grade cervical intraepithelial neoplasia group,low-grade cervical intraepithelial neoplasia group,and normal cervical group were 96.63%(86/89),72.25%(125/173),34.48%(20/58)and 6.06%(4/66),respectively.All the four groups and pairwise comparisons between the groups showed statistical differences(P<0.0001).ROC curve analysis SOX1 gene methylation has high sensitivity and specificity in the diagnosis of cervical cancer with 96.63% and 95.45%,respectively,and the area under the curve is 0.96.3.In the exfoliated cells of cervical lesions,the positive rates of SOX1 gene methylation in the cervical cancer group,High-grade squamous intraepithelial lesion group,Low-grade squamous intraepithelial lesion group,and non-lesion group were 97.92%(47/48),62.50%(70/112),23.61%(34/144)and 17.20%(43/250),respectively.Pairwise comparisons between the four groups and groups,except for the low-grade group and the disease-free group,there was no statistical difference(P=0.1222),and the differences in the other groups were statistically significant(P<0.0001).ROC curve analysis SOX1 gene methylation has a high sensitivity and specificity of 93.75% and 97.20% in the diagnosis of cervical cancer,and the area under the curve is 0.965.Conclusions:1.In cervical lesion tissue and cervical lesion exfoliated cells,the SOX1 gene in cervical cancer group has hypermethylation,and its positive rate of methylation increases with the severity of cervical lesions.2.SOX1 gene methylation has high sensitivity and specificity for the diagnosis of cervical cancer,and it can be a potential molecular marker for early screening of cervical cancer. |
