Objective: To investigate the correlation between methylation of PAX1,SOX1 and ZNF582 genes and oral carcinogenesis in oral cancer tissues.Methods: We collected the surgical patients who were diagnosed and treated in in the first people's Hospital of Xiangtan and Xiangtan Central Hospital from January to December,2016.Specimens consist of 10 cases of oral mucosal benign lesions and 30 cases of oral cancer.All of oral cancer patients were diagnosed as oral squamous cell carcinoma by clclinical pathomorphology,and the specimens was detected in the tumor tissue,para cancer tissue and surgical margin tissue after radical resection of oral cancer.And all cases were not treated with antitumor treatment before operation.Methylation specific real-time fluorescence quantitative polymerase chain reaction(QMSP)was used to detect the methylation level of PAX1,SOX1 and ZNF582 genes.1.Comparison of benign and malignant oral lesions in PAX1,SOX1 and ZNF582 gene methylation differences;2.Compare the differences of the oral cancer patients PAX1,SOX1 and ZNF582 gene methylation between cancer tissues,para cancer tissues and between surgical margin tissues.The prognosis of these 40 patients was continuously traced.Correlation analysis of patients with oral cancer PAX1,SOX1 and ZNF582 gene methylation the clinical and pathological data and prognosis.Results:1.The methylation level index(M-index)and the positive rate of methylation of PAX1,SOX1 and ZNF582 gene in oral cancer tissues were significantly higher than those in benign oral lesions(P < 0.001).2.The methylation level index of PAX1 / SOX1 gene was higher in cancer tissues than that in paracancerous tissues(P < 0.05),and the positive rate of ZNF582 methylation in cancer tissues was higher than that in paracancerous tissues(P <0.05).3.There was no significant difference between the methylation of PAX1,SOX1 and ZNF582 gene and the age,sex,location,tumor size,clinical stage and pathological differentiation type of patients.4.Patients with PAX1,SOX1 and ZNF582 gene methylation positive was shorter than that of methylated negative patients,but there was no statistical significance(P=0.115?0.684?0.493).Multivariate Cox regression was used to analyze methylation status,age,sex.There was no significant difference in histological differentiation,lymph node metastasis,pathological stage and prognosis(P>0.05).Conclusion:1.Hypermethylation of PAX1,SOX1 and ZNF582 genes are correlated with oral cancer.2.The methylation of PAX1,SOX1 and ZNF582 genes in cancer tissues is higher than that in adjacent tissues and margin tissues,which is great significance in the diagnosis of oral cancer.
