Objective: To investigate the effects of propofol on the expression level of CXCR4/CXCR7 and its migration ability of human breast cancer MCF-7 cells in vitro.Methods: MCF-7 cells were randomLy divided into 4 groups(group C,group E,group P1 and group P2)for treatment with basal medium,lipid emulsion,or propofol at 3 ?g/mL and 8 ?g/mL.The viability was measured by MTT assay.The migration ability was detected by wound-healing assay and transwell assay.The mRNA expression level of CXCR4/CXCR7 was detected by RT-qPCR;The protein expression level of CXCR4/CXCR7 was determined by Western blot.Results: Compared with group C,the scratching healing rates in group P1 and P2 were decreased(20.01±1.48 %,18.36±1.80 %,P<0.05),and the chemotactic index was also decreased(1.09±0.06,1.00±0.05,P<0.05).Western blot showed the protein expression level of CXCR4/CXCR7 was decreased in group P1 and P2.However,both the mRNA expression level of CXCR4/CXCR7 and the viability of the MCF-7 cells were kept no change.Conclusion: Propofol down-regulates the protein expression levels of CXCR4/CXCR7 and inhibits the migration ability of breast cancer MCF-7 cells in vitro.
