Objective:Piglet brain injury model was used to verify the therapeutic effect of intralaminar hypothermia in craniocerebral injury,and to further explore the possible mechanism.Methods:1.Five male piglets weighing about 15 kg were selected for the preliminary experiment of brain injury model.2.Relying on the above modeling method,12 male piglets weighing about 15 kg were re-selected for modeling.After that,the model was randomly divided into experimental group and control group,with 6 piglets in each group.The control group was given normal saline at room temperature,while the experimental group was given low-temperature saline infusion at 4 degrees Celsius3.Anal temperature,blood sugar and arterial blood PH were measured at different time points in two groups.4.Behavior of two groups of piglets was observed and their neurological function was evaluated.5.Lumbar puncture was used to monitor the changes of intracranial pressure at different time points in the two groups.6.The changes of CT results in the experimental group and the control group were observed by CT scanning at different time points.7.The brain tissues of 1 cm around craniocerebral injury in the experimental group and the control group were taken for pathological study.8.SPSS 22.0 software package was used for statistical analysis of all experimental data.Results:1.The model of brain injury in piglets was established successfully.2.Relying on the above modeling method,12 male piglets weighing about 15 kg were re-selected for modeling.After that,the model was randomly divided into experimental group and control group,with 6 piglets in each group.* The control group did not add any intervention measures.The experimental group established a transfusion route for skull plate barrier,and then was treated with hypothermic infusion of 4 ?.3.Compared with the control animal model,there was significant difference in neurological function score in the intraplate hypothermia group(P < 0.05).4.Compared with the control group,the intracranial pressure in the intraplate hypothermia group was significantly lower(P < 0.05).5.Compared with the control animal model,the CT results of intraplate hypothermia group showed that the injury area was reduced and the hematoma absorption was obvious,the difference was statistically significant(P < 0.05).6.Compared with the control animal model,HE pathological results showed that the inflammatory cells around the brain injury tissue decreased and the pathological changes of brain tissue were significantly alleviated in the intraplate hypothermia treatment group.Conclusions:1.The preparation of piglet animal model is stable,the success rate is high,and it is easy to survive.It can pave the way for the next experiment.2.Mild hypothermia can reduce intracranial pressure,reduce the metabolic state of brain cells,alleviate inflammatory reaction of brain tissue,regulate coagulation function,improve the quality of life of patients,reduce mortality and disability rate,and effectively improve the prognosis of severe craniocerebral trauma animals.3.Temperature reduction within the skull plate barrier is effective in the treatment of craniocerebral injury in piglets.
