Objective:By detecting the expression of Raf,Spry and HPV in cervical tissues at different levels of pathological changes,exploring the function between Raf and Spry in the evolution of cervical squamous cell carcinoma,and the the correlation of each others.Methods:133 cases of cervical tissues were collected from October 2015 to December 2017 in the Department of Obstetrics and Gynecology,First Affiliated Hospital of Henan University of Science and Technology,including 50 cases of cervical squamous cell carcinoma,32 cases of highly squamous intraepithelial lesions,22 cases of low grade squamous intraepithelial lesions,29 cases of normal cervical tissue,and every tissue was confirmed by pathology.HPV test of cervical tissue was carried out for all patients before sampling.The expression of Raf and Spry in 133 cases of cervical tissues were detected by immunohistochemistry.The relation between the expression of Raf and Spry with the clinical pathology character of cervical squamous cell carcinoma was measured.The correlation of the expression of Raf and Spry in cervical squamous cell carcinoma was also assessed,also the correlation of the expression of Raf,Spry and HPV in cervical squamous cell carcinoma can be measured.Results:1.The positive rate of Raf protein in normalcervical tissue,low-grade squamous intraepithelial lesion,high-grade squamous intraepithelial lesion and cervical squamous cell carcinoma increased gradually.The differences in comparison between any two groups are all statistically significant(P<0.05).The positive rate of Spry protein in normalcervical tissue,low-grade squamous intraepithelial lesion,high-grade squamous intraepithelial lesion and cervical squamous cell carcinoma decreased gradually.The differences in comparison between any two groups are all statistically significant(P<0.05).2.Raf is associated with clinical stage(FIGO in 2009,?-? period,?-? period)and lymphatic metastasis(no transfer,transfer)in cervical squamous cell carcinoma(P<0.05),but it didn't relate to age(age<50years,age?50years)and pathological grade(high,medium-low differentiation)(P>0.05).When it occurs high clinical stage or lymphatic metastasis,the positive rate of Raf protein would be higher.Spry is associated with pathological grade of cervical squamous cell carcinoma(P<0.05),but it didn't relate to age,clinical stage and lymphatic metastasis(P>0.05).The positive rate of Spry protein would be lower with low degree of differentiation.3.Raf protein was positively correlated with HPV infection in cervical lesions(P<0.05);Spry protein was negatively correlated with HPV infection(P<0.05);but negatively correlated with Raf and Spry protein expression(P<0.05).Conclusions:1.With the aggravation of cervical lesions,the positive rate of Raf protein gradually increased,while that of Spry protein gradually decreased,which showed it may be related to the occurrence of cervical squamous cell carcinoma.2.In cervical squamous cell carcinoma,the expression of Raf protein Was related to clinical stage and lymphatic metastasis(P<0.05),while Spry protein is related to pathological grade(P<0.05),which suggests that Raf and Spry proteins may be involved in the early development,metastasis and prognosis of cervical squamous cell carcinoma.3.Raf and Spry proteins may be involved in the evolution of cervical squamous cell carcinoma caused by HPV infection.
