Abstract/Summary:
Objective:Detecting the expression of connective tissue growth factor (CTGF) protein and transforming growth factor-β1(TGF-β1) protein in normal cervical tissue, CIN Ⅱ-Ⅲ and cervical squamous cell carcinoma tissues by immunohistochemistry. Then investigate the relationship between the expression of these two kinds of proteins in cervical squamous cell carcinoma and the clinical pathological features, the pelvic lymph node metastasis especially.Methods:1.10cases of normal cervical tissue,20cases of CIN Ⅱ-Ⅲ and98cases of cervical squamous cell carcinoma tissues were used to detect the expression of connective tissue growth factor (CTGF) protein and transforming growth factor-β1(TGF-β1) protein by immunohistochemistry SP method. Then analysis the correlation between the expression levels of these two proteins in cervical squamous cell.2. To investigate the relationship between the expression of CTGF and TGF-β1protein and the clinical pathological features of cervical squamous cell carcinoma.3. To explore the relationship between the expression of CTGF and TGF-β1protein in cervical squamous cell carcinoma and the pelvic lymph node metastasis 4. Date analysis using SPSS18.0software. Level of significance was set at α=0.05.Results:1. The expression of CTGF and TGF-β1protein were mainly localized in the cytoplasm and cell membrane, present yellow or brown. The positive expression rates of two proteins in the normal cervix, CIN Ⅱ-Ⅲ, cervical squamous cell carcinoma tissue were10.0%,40.0%,80.6%;20.0%,45.0%,62.5%respectively and a trend of increasing has been observed, the difference between three groups have statistically significant (P=0.000<0.05). The expression of CTGF and TGF-β1protein in cervical squamous cell carcinoma was positively correlated (r=0.643,p=0.000<0.05);2.The positive expression of CTGF and TGF-β1protein in cervical squamous cell carcinoma were related to differentiation of cervical squamous cell carcinoma, lymph-node involvement and clinical stages (P <0.05), however not related to age, vascular invasion, tumor diameter, and depth of myometrial invasion;3. Single factor analysis was performed between cervical squamous cell carcinoma group with and without pelvic lymph node metastasis. The expression of CTGF and TGF-β1proteins, age, Depth of myometrial invasion, vascular invasion, tumor diameter and degree of differentiation are related to pelvic lymphatic metastasis of cervical squamous cell carcinoma (P<0.05), further logistic regression analysis showed that CTGF protein, age, histological grade and tumor diameter were the independent risk factors of Pelvic lymphatic metastasis of cervical squamous cell carcinomaConclusion:1. Comparing with normal cervical tissues and CINⅡ-Ⅲ tissues, the expression of CTGF and TGF-β1were up-regulated in cervical squamous cell carcinoma tissues, and the expression of the two proteins was positively correlated. It reveals that both CTGF and TGF-β1may take part in the process of cervical squamous cell carcinoma.2. There was a significant association between expression of CTGF and TGF-(31with differentiation of cervical squamous cell carcinoma, lymph-node involvement and clinical stages. It presents that both CTGF and TGF-β1may play roles in the invasion and metastasis of cervical squamous cell carcinoma. High level CTGF maybe an independent risk factor of pelvic lymphatic metastasis of cervical squamous cell carcinoma.
