| ObjectiveTo observe the inhibited proliferation and apoptotic induction effects of berberine on human ovary cancer line OVCAR3,and explore the potential value in the ovarian cancer treated with berberine.MethodsOVCAR3 cells in logarithmic phase were treated with 50μmol/L,100μmol/L,200μmol/L,and 500μmol/L.No drugs were added to the control group.The CCK8 method was used to measure proliferation influences of OVCAR3 cells.The cell apoptosis rates were detected by flow cytometric.The mRNA and protein expressions of Caspase-3,Caspase-8 in the ovarian cancer cells treated with the berberine were tested by RT-PCR and Western blotting.The t-test comparison analysis was adopted expression difference of Caspase-3,Caspase-8 among groups.Results1.The test results by CCK-8 are shown:OVCAR3 cells were treated with different concentrations of berberine for 24h,the growth inhibition rates were increased to(32.20±1.94)%for 50μmol/L group,(50.97±0.65)%for 100μmol/L group,(69.53±1.09)%for 200ltmol/L group,(79.53土0.08)%for 500μmol/L group(compared with the 50μmol/Lgroup,P=0.000 for the other group).Difference was statistically significant.The growth inhibition rates were increased to(50.97±0.65)%,(83.89±0.82)%for 48h group,(93.06±0.33)for 72h group after treated with 1 00μmolmol/L group(compared to the 24h group,P values were 0.000 for the other group).Berberine could effectively inhibit the proliferation of OVCAR3 cells in a time-and dose-dependent manners.2.The test results by flow cytometry are shown:After OVCAR3 cells treated with berberine with the optimal concentration(100μmol/L)and the optimal work time(24h),the apoptotic ratio of cell in the experiment group increased significantly compared with the control group(4.2010.26%VS 50.9310.40%),and the difference between them showed statistically significant(P<0.01).3.The test results by RT-PCR are shown:the expression levels of apoptosis-related Caspase-3 mRNA(1.63±0.14 VS 1),Caspase-8 mRNA(1.76±0.14 VS 1)in the group treated with berberine increased significantly,compared with the control group,and the difference between them showed statistically significant(P=0.001).4.The test results by Western bolt are shown:the expression levels of apoptosis-related proteins Caspase-3(activated)(0.389±0.011 VS 0.099±0.003),Caspase-3(0.570±0.014 VS 0.276±0.007),Caspase-8(activated)(0.386±0.011 VS 0.3 18±0.004),Caspase-8(0.571±0.015 VS 0.394±0.014)increased significantly,compared with the control group,and the difference between them showed statistically significant(P<0.001).ConclusionThe berberine can inhibit proliferation and induce apoptosis of human ovary cancer line OVCAR3 with anti-tumor effects,which provide the experimental basis and theory support for ovarian cancer treatment clinically with the berberine... |
