To Study The Effects Of Berberine On The Expression Of SKOV-3?A2780 TDO,AhR MRNA In Human Ovarian Cancer Cells

Background and objectives:Ovarian cancer(OC)is the highest mortality of female genital malignant tumor.Immunologic escaping is one of the main factors leading to the poor prognosis of OC patients.Tryptophan-2,3 dioxygenase(TDO),is the key factor which mediates the immunodepression and tumor immunologic escaping in ovarian carcinoma.Inhibiting the expression or the activity of TDO can improve the immunity of ovarian cancer patients,and ameliorate the prognosis.Berberine can inhibit the activity of TDO,but if berberine can down regulated the expression of TDO and aryl hydrocarbon receptor(Ah R)is lack of research.This experiment will study the effects of berberine on the expression of TDO,Ah R mRNA and the activity of TDO in human ovarian cancer cells.Methods:Using RT-PCR to detect SKOV-3 and A2780 cells TDO,AHR mRNA expression after the effects of berberine with different concentrations at different time?Results:1?Our study found that A2780 and SKOV-3 have high expression of TDO;2?Berberine can inhibite the growth of SKOV-3 and A2780 cells in vitro(IC50 are2.48±0.269?mol/L and 2.41±0.157?mol/L,respectively);3?Berberine could significantly inhibit the expression of AHR mRNA in the SKOV-3 cells,and the inhibitory activity increasing with drug concentration and the time enhancement,while berberine concentration dependently reduced SKOV-3 cell supernatant of tryptophan depletion and kynurenine production;4?Berberine could not significantly inhibit the expression of AHR mRNA in the A2780 cells.Conclusion:1?Our study found that A2780 and SKOV3 are high expression of TDO;.2?Berberine down-regulates TDO mRNA expression and may reduce the tryptophan depletion of ovarian cancer cells,thereby decreasing kynurenine metabolism and down-regulating AHR expression.3?Berberine can overcome the immunodepression of ovarian cancer,and maybe,could well prove effective in both of type I and type II ovarian cancer.
