Background and objectives: Ovarian cancer is one of the most common death causes of various malignant tumors in gynecology because of various difficulties faced by clinical treatment.PD-1(programmed death,programmed cell death)and PD-L1(programmed cell death-Ligand 1,cell death-ligand 1),both of which can synergistically inhibit the body's immune function,they belong to the B7 family,This family is an important co-stimulatory molecule in the body,and its members' receptors and ligands can play a common role in the proliferation of T cells and the production of cytokines.In our laboratory,it has been demonstrated that the concentration of the berberine extract has a concentration-and time-dependent inhibitory effect on human ovarian cancer cells.This study aims to investigate the effect of berberine on the human ovarian cancer cell from the perspective of immunotherapy for ovarian cancer.-Expression of PD-L1 mRNA.Methods: 1,Take berberine at an IC50 concentration 72 hours after acting on human ovarian cancer cells A2780 and SKOV-3 to add two cell lines;2,The two groups of cell lines in the experimental group were treated with berberine under IC50 concentration for 2 h,6 h;3,The expression of PD1 and PD-L1 mRNA in A2780 cells and SKOV-3 cells after berberine treatment by polymerase chain reaction(RT-PCR).Results: 1.The berberine with IC50 concentration of 72 h after acting on human ovarian cancer cells A2780 and SKOV-3 was added to two cell lines,respectively;2.The two groups of cell lines in the experimental group were treated with berberine at an IC50 concentration for 2 hours and 6 hours;3.The expression of PD1 and PD-L1 mRNA in A2780 cells and SKOV-3 cells after berberine treatment was detected by polymerase chain reaction(RT-PCR).Conclusion: 1?Human ovarian cancer cells express PD-L1,which may be related to the immune escape of ovarian cancer;2,Berberine can inhibit the transcription of PD-L1 mRNA,and the inhibitory effect is positively correlated with the action time;3,Berberine may improve the immunosuppressive state of ovarian cancer,inhibit the immune escape of ovarian cancer,and may improve the prognosis of the ovary.
