Objective:A polycystic ovary syndrome(PCOS)rat model was established and glutamine intervention was given to investigate the regulatory effect and optimal concentration of glutamine on inflammatory state and oxidative stress in PCOS rat model,and to provide new ideas and theoretical basis for the prevention and treatment of PCOS.Methods:PCOS rats model were constructed by subcutaneous injection of dehydroepiandrosterone(DHEA),and the control group was injected with the same amount of oil;PCOS model rats were divided into the following four groups according to the dose of Gln: PCOS group,PCOS+0.5g/kg Gln group,PCOS+0.75g/kg Gln group,PCOS+1.0g/kg Gln group.Gln was administered by intraperitoneal injection in the next morning after modeling,and PCOS group was given the same amount of saline.The rats were administered again at intervals of 4 h,and the rats in each group were sacrificed 24 h after the first intraperitoneal injection,and blood and ovarian tissues were collected.The main observation indexes were as follows:1.Body weight and bilateral ovarian weight of rats in each group were weighed;2.Morphological analysis of ovarian tissue by HE staining;3.Serum estradiol(E2),progesterone(P),testosterone(T),luteinizing hormone(LH)and follicle-stimulating hormone(FSH)were detected.4.Fasting insulin and blood glucose were measured at the level of insulin resistance,and HOMA-IR was calculated;5.Serum interleukin-6(IL-6),interleukin-18(IL-18),C-reactive protein(CRP)and tumor necrosis factor-a(TNF-a)were detected at the level of inflammatory factors,and the expression levels of these four inflammatory factors in ovaries were detected by immunohistochemical staining;6.Serum superoxide dismutase(SOD),nitric oxide synthase(NOS),malondialdehyde(MDA)and nitric oxide(NO)were detected at the level of oxidative stress.Results:1.Comparison results of the observation indexes of PCOS group and control group:The ovarian morphology of PCOS group showed ovarian polycystic changes.Compared with the control group,the body weight,bilateral ovarian weight,serum T and LH levels,Serum INS,HOMA-IR,serum and ovarian IL-6,IL-18,CRP,TNF-a levels,serum MDA,NO,NOS levels in PCOS group were significantly increased,while SOD levels were significantly decreased,the differences were statistically significant.2.Comparison results of Gln intervention groups and its comparison with PCOS group:(1)There was no significant change in morphological changes of ovarian tissue.(2)Compared with the PCOS group,body weight,bilateral ovarian weight,serum E2,P,LH,FSH levels,blood FBG,INS,HOMA-IR in Gln intervention groups were not significantly different;Comparing the Gln intervention groups in pairs,there were also no significant difference in these indicators.(3)Comparisons of inflammatory status:Compared with the PCOS group,the levels of IL-6,IL-18,CRP and TNF-a in the serum and ovarian tissue in PCOS+0.5g/kg Gln group and the PCOS+0.75g/kg Gln group decreased significantly,while there was no significant difference in these inflammatory factors in PCOS+1.0g/kg Gln group.Comparing the Gln intervention groups in pairs,The levels of inflammatory factors in the serum and ovary in the PCOS+0.5g/kg Gln group and the PCOS+0.75g/kg Gln group were significantly lower than those in the PCOS+1.0g/kg Gln group.There was no significant difference of the levels of inflammatory factors in the serum of the two groups,but the levels of inflammatory factors in the ovary in the PCOS+0.75g/kg Gln group were lower than those in the PCOS+0.5g/kg Gln group,and the difference was statistically significant.(4)Comparison of oxidative stress status:Compared with the PCOS group,the levels of MDA,NO and NOS in the PCOS+0.5g/kg Gln group and PCOS+0.75g/kg Gln group were significantly decreased,and the levels of SOD were significantly increased,while the levels of MDA,NO and SOD in the PCOS+1.0g/kg Gln group were not significantly different.Comparisons of the Gln intervention groups in pairs showed that there was no significant difference in the level of oxidative stress between the PCOS+0.5g/kg Gln group and the PCOS+0.75g/kg Gln group.In addition to NOS,other indicators in the two groups were better than those in the PCOS+1.0g/kg Gln group.Conclusions:1.PCOS rat model was successfully constructed in this study.2.The levels of inflammatory cytokines IL-6,IL-18,CRP and TNF-a in PCOS rats were abnormally increased,and Gln could alleviate the inflammatory state of PCOS rats,with the optimal concentration of 0.75g/kg.3.The oxidative stress state in PCOS rats was enhanced,and Gln could alleviate the oxidative stress state in PCOS rats,and the optimal concentration was 0.75g/kg.
