Objective:It is of great practical significance to observe and evaluate the mechanism of the MAPK pathway in the treatment of ERK1,ERK2 and P38 proteins in SANFH rabbits by chuan gu pian,a hospital preparation,so as to explore a better therapeutic method,further improve the prevention and early treatment of this disease,and maximize the effect of improving the patient's own joint function.Methods:40 healthy male Japanese big eerian rabbits,weighing 2.0-2.5kg,were provided by experimental center of chengdu university of traditional Chinese medicine(license no.: no.99011,chuan shi dong guan),qualified for quarantine use.This study animals and animal experiments are completed in the center of chengdu university of traditional Chinese medicine experiment,experimental animal for femoral head and abdominal aorta collected blood,drawn by air embolism method executed after experimental animals.Animal grouping: 40 Japanese male big-eared rabbits were randomly divided into 2 groups,namely the blank group(12 rabbits)and the model group(28 rabbits).After successful modeling,8 blank rabbits and 24 model rabbits remained.Model group of rabbits were randomly divided into model control group,sichuan and bone treatment group,a fairy bone capsule treatment group,the eight only.Observation indexes :(1)the appearance,morphology and color of articular cartilage of femoral head were observed by naked eye,and bone hardness was felt by cutting with a knife.(2)HE staining,under light microscopy to observe the changes of the morphological structure of the femoral head.(3)electron microscopy fixed liquid handling,sliced paraffin embedding,in femoral head ultrastructure change observed under transmission electron microscope.(4)the contents of p-ERK1,p-ERK 2 and p-P38 proteins were detected by Western blotting.Results:1.Through statistical analysis,blank group rabbit bone empty pit rate was(14.31 + 1.55)%,the model group rabbit bone empty pit rate was(56.07 + 2.14)%,the differences between the two groups have statistical significance(P < 0.01).2.Immunohistochemical: after the success of the building,compared with the blank group,model group rats P38 lightning protein expression significantly raised,six weeks after treatment,the treatment group compared with control group P38 lightning protein expression were significantly reduced,and sichuan bone treatment group compared with the faeries bone remains capsule treatment group P38 lightning protein expression no statistical difference(P > 0.05).3.APK signal transduction-related protein expression: the expression of MAPK signaling pathway related proteins ERK1 and ERK2 was analyzed and detected by Western blot method,and it was found that the expression of p-ERK 1 and p-ERK 2 in the control group was significantly down-regulated compared with the blank group.The treatment group compared with control group,p-ERK1,p-ERK2 expression were significantly increased.Meanwhile,there was no significant difference in the expression of p-ERK 1 and p-ERK 2 between chuangupian treatment group and xianling gubao capsule treatment group(p >0.05).Conclusion: 1? Hormones can down-regulate the expression of ERK1 and ERK2 proteins,and promote the occurrence and development of avascular necrosis of femoral head.Chuangu tablets can promote the expression of ERK1 and ERK2 proteins,inhibit the fat production of femoral head bone marrow cells,improve femoral head microcirculation,reduce hip cavity pressure,and promote the growth and function of osteoblasts.Can effectively restrain the development of ischemic necrosis of femoral head,will exert good effects on treatment of early avascular necrosis of femoral head.2? Hormone by raising P38 lightning protein expression,accelerate bone cells apoptosis,eventually lead to femoral head necrosis.The active components of traditional Chinese medicine chuangu tablet can also antagonize the effects of hormones through signal pathway and produce the effect of reverse regulation.By cutting P38 lightning protein gene expression,can slow down the bone cells apoptosis,reduce the incidence of femoral head empty bone pit.Chuan gu pian can promote the repair and reconstruction of necrotic osteocytes,inhibit the apoptosis of osteocytes,and play an early role in the treatment of steroid-induced femoral head necrosis.3? Chuangu tablets can promote the osteogenic differentiation of bone marrow mesenchymal stem cells,and is conducive to the repair of necrotic femoral head tissue.The therapeutic effect of chuan gu tablet on steroid-induced avascular necrosis of femoral head can activate MAPK signaling pathway and regulate the expression of related factors ERK1,ERK2 and P38,thereby inhibiting the apoptosis of bone marrow stromal cells and promoting the repair and reconstruction of osteoblasts.
