Objective:To determine the expression levels of RNA binding motif protein 8A(RBM8A,Y14)and?II spectrin(beta2 Spectrin,SPTBN1,?2SP)in tumor tissues and paracancerous tissues of patients with colorectal carcinoma(CRC),to preliminarily analyze the relationship between the two proteins and clinical data and explore the correlation between RBM8A and SPTBN1,so as to further explore the expressions of the two in CRC and their significance.Methods:1.Using UniHI and ONCOMINE bioinformatics database,it was found that RBM8A and SPTBN1 were differentially expressed in CRC tissues and paracancerous tissues.A literature review was conducted correspondingly,to explore the expression of target proteins RBM8A and SPTBN1 in CRC.2.The postoperative CRC tissues and corresponding paracancerous tissue specimens from patients who were treated in the First Hospital of Qinhuangdao City were collected as the experimental group(n=60)and the control group(n=60)respectively.3.Immunohistochemistry(IHC)and Western blot were used to detect the expression of RBM8A and SPTBN1 in CRC tissues and paracancerous tissues.4.Statistical methods:SPSS21.0 statistical software was used for statistical analysis and histogram drawing.Western blot results were expressed as mean±standard deviation(x±s),and IHC results were expressed as a percentage.(1)We analyzed whether there was differential expression of RBM8A and SPTBN1 in CRC tissues and paracancerous tissues:Quantitative data were analyzed by t-test,and qualitative data were analyzed by?2 test or Fisher exact probability method.(2)The relationship between the RBM8A,SPTBN1 expressions in CRC tissues and the clinical data was analyzed:using?~2 test,Fisher exact probability method,nonparametric test based on rank,nonparametric rank correlation analysis and Logistic regression analysis.(3)The correlation between RBM8A and SPTBN1 expression in CRC tissues was analyzed:Nonparametric rank correlation analysis was performed.The difference was statistically significant at the test level?=0.05,and P<0.05.Results:1.Results of RBM8A experiment(1)Western blot results showed that RBM8A was expressed in CRC tissues and paracancerous tissues,however the difference was not statistically significant(t=0.974,P=0.332).(2)IHC results1)RBM8A was expressed in CRC tissues and corresponding paracancerous tissues,and the difference was statistically significant(?~2=11.109,P=0.001).2)There was significant difference in RBM8A expression level in terms of differentiation(?~2=16.367,P=0.000),tumor diameter(?~2=8.910,P=0.003)and serosal infiltration(?~2=13.806,P=0.000)grouping.3)The expression of RBM8A was negatively correlated with the degree of differentiation(r=-0.460,P=0.000),and was positively correlated with tumor diameter(r=0.575,P=0.000)and the degree of serosal infiltration(r=0.620,P=0.000).4)Degree of differentiation(OR=5.900,95%CI:2.035-17.104,P=0.001),tumor diameter(OR=6.429,95%CI:1.757-23.524,P=0.005),and degree of serosal infiltration(OR=10.200,95%CI:2.689-23.693,P=0.001)were all associated with the expression of RBM8A.2.Results of SPTBN1 experiment(1)Western blot result indicated that SPTBN1 was expressed in CRC tissues and paracancerous tissues,and the difference was statistically significant(t=-2.662,P=0.010).(2)IHC results1)SPTBN1 was expressed in CRC tissues and corresponding paracancerous tissues,the difference was statistically significant(?~2=11.442,P=0.001).2)The difference of SPTBN1 expression level was statistically significant in the differentiation(?~2=10.048,P=0.007),tumor diameter(?~2=4.077,P=0.043)grouping.3)The expression of SPTBN1 was positively correlated with the degree of differentiation(r=0.406,P=0.001),but was negatively correlated with tumor diameter(r=-0.261,P=0.044).4)Degree of differentiation(OR=0.282,95%CI:0.122-0.653,P=0.003)and tumor diameter(OR=0.341,95%CI:0.118-0.982,P=0.046)were all associated with the expression of SPTBN1.3.The expression of RBM8A was negatively correlated with SPTBN1 in CRC(r=-0.267,P=0.039).Conclusions:1.The expression level of RBM8A in CRC tissues was higher than that in paracancerous tissues,which was related to the degree of differentiation,tumor diameter and presence of serosal infiltration,suggesting that RBM8A has a certain relationship with the degree of malignancy,proliferation and invasion ability of CRC.2.The expression level of SPTBN1 in CRC tissues was lower than that in paracancerous tissues,which was related to the degree of differentiation and tumor diameter,indicating that SPTBN1 had a certain relationship with the malignant degree and proliferation ability of CRC.3.In CRC tissues,the expression of RBM8A was negatively correlated with SPTBN1,suggesting that there might be mutual regulation relationship between the two proteins and that they both act on the occurrence and development of CRC.
