Objective:Detect the expression of RBM8 A in colorectal carcinoma,and explore its clinical features in human colon cancer.Methods:1.Some specimens were obtained form radicaloperation for carcinoma of colon and pathological diagnosis of CRC.qRT-PCR?Western blot and immunohistochemistry analysis were used to examined the expression of RBM8 A in these tissues and adjscent tissues.Collected the clinical and pathological data of these CRC patients,then analysisd the correlation bet-ween RBM8 A and CRC clinicopathologic features.2.Collectting some serum specimens of healthy person and patients with colorectal cancer.Using immunohistochemistry to analyzethe the expr-ession of RBM8 A in these specimens.3.Culture the normal human colon epithelial cell NCM460?human colon cancer cell lines SW480?SW620?HT29?LOVO?T84 and COLO320 DM respectively.qRT-PCR and Western blot were used to examined the expression of RBM8 A in these seven cell lines.Results:1.qRT-PCR results of tissues showed that compare with the tumor-ad-jacent tissues,the relative expression of RBM8 A mRNA in colorectal car-cinoma tissues is 1.256±0.473,P<0.000.The difference was statistically significant.The expression of RBM8 A mRNA in colorectal cancer was no-t significantly correlated with gender,age,CEA level,lymph node metast-asis,tumor differentiation or TNM stage.Western blot results shows that RBM8 A protein was expressed in both tumor-adjacent tissues and colorectal carcinoma tissues.The gray-scalev alue statistics results are 1.637±0.885 and 0.9824±0.152,P<0.000.The dif-ference was statistically significant.The expression of RBM8 A protein in these colorectal cancer tissues was not significantly correlated with gende-r,age,CEA level,lymph node metastasis,tumor differentiation or TNM stage.The immunohistochemical results showed that RBM8 A was highly ex pressed in some colorectal cancer tissues,and the protein was mainly loca-ted in the cytoplasm and nucleus,with brown-yellow or brown staining.According to our scoring methods,there was no obvious expression in the cytoplasm of adjacent tissue.The positive expression rates of RBM8 A in colorectal cancer tissues and tumor-adjacent tissues were respectively 24% and 0,P<0.05.The difference was statistically significant.The expression of RBM8 A in these colorectal cancer tissues was not significantly correlated with gender,age,CEA level,lymph node metastasis,tumor diff-erentiation or TNM stage.2.ELISA results of serum specimens showed that the expression of RBM8 A in colorectal cancer patients was much higher than in healthy per-son.The concentration of RBM8 A were 4.467±1.371 and 2.585±0.923 res-pectively,The difference was statistically significant.3.qRT-PCR results of cell lines showed that compare with the normal human colon epithelial cell NCM460,the expression of RBM8 A in oth-er six colon cancer cell lines were COLO320DM:8.75±0.30;SW620:3.78±0.23;LOVO:1.47±0.61;SW480:1.43±0.16;HT29:1.38±0.12;T84:1.11±0.03.And there was no significant difference between the other cell lines and NCM460.The difference was statistically significant.According to the results of Western blot,RBM8 A protein was expres-sed in all of seven cell lines.The gray-scale value statistics showed that there was a significant higher expression of RBM8 A protein in SW620 a-nd COLO320DM(P < 0.05),The difference was statistically significant,while there was no significant difference between the remaining cell lines and the normal cell lines.Conclusion:The expression of RBM8 A is increased in colorectal cancer tissues and serums.Its potential value can be further explored.
