Background and object:Colorectal cancer(CRC)is a common cancer of the digestive tract,including the rectum and colon.For the present,the most common reason for the occurrence of cancer in the large intestine is the gradual increase of age.Diet: high in fat and low in fiber.What is more worrying is that most of colorectal patients do not have typical and obvious symptoms in the early onset of occult Colorectal cancer,which makes the diagnosis of colorectal cancer more delayed than other cancers.Early detection and diagnosis are the key to improve the efficacy and prognosis of colorectal cancer.Proteomics is a large-scale study of proteins,especially the structure and function of proteomics.Protein is an important component of an organism because it plays a central and key role in the metabolic pathway of cell physiology.Proteomics has been favored by researchers for its advantages in quantitative detection and differential identification of a large number of proteins in samples in a short period of time.This study collected the colorectal cancer tissue and corresponding normal tissue health,analyze detection using the above methods,eventually capture expression protein 25.Among 25 differentially expressed proteins are 18 relatively high expression proteins including KRT18 and 7 relatively low expression proteins including TPM2.Immune mark identification technology was further used to identify proteins with significant difference such as protein 2(anterior gradient-2,AGR2),immunohistochemical technique was used to analyze clinical pathologic features.aims to explore the pathological characteristics and the colon cancer occurrence,development and transfer.Methods:The following three parts discuss the main contents of this study:1.Two-dimensional liquid chromatography and mass spectrometry technology were used to detect tissue samples(large intestine cancer tissue and corresponding normal healthy tissue),relatively high and low proteins expression were obtained from the specimen.Methods:(1)Polypeptides were prepared from the total protein extracted from the paired tumor tissues collected by pyrolysis.(2)Protein species and expression levels in paired tissues are determined by chemical analysis and mass spectrometry combined with two-dimensional liquid chromatography.(3)Statistical analysis was performed on the expression levels of different proteins identified in the above steps to obtain the list of differentially expressed proteins.2.Detect AGR2 protein expression levels in the paired tumor tissue: Select four groups of colorectal adenocarcinoma tissue samples from different tumor TNM staging.Western Blot is used to observe whether AGR2 protein in colorectal adenocarcinoma tissues have a relatively high expression.3.Analysis of the potential association between the difference in general clinical data of patients with colorectal cancer and the relatively high and stable expression of differentially expressed protein AGR2 in the patient's tumor tissue: Collect colorectal cancer tissue samples from different TNM stages,ages and gender.Observe the expression of the target protein AGR2 using immunohistochemical method on sections of specimens.Statistical analysis and comparison method were performed on patient's gender,age and other data in combination with the expression of the target protein.Results:1.25 proteins with statistically significant differencein expression were identified: 18 relatively high expression proteins including KRT18 and 7 relatively low expression proteins including TPM2.2.Western Blot results confirmed that the expression level of AGR2 in colorectal cancer tissues was significantly higher than that in adjacent tissues,which further validated the results of the first part of the two-dimension liquid chromatography combined with mass spectrometry.3.(1)The results of immunohistochemical staining shows high expression of AGR2 protein in colorectal cancer tissues ismainly in cytoplasm.(2)The expression of AGR2 protein is correlated with TNM staging,differentiation degree and distant metastasis of colorectal cancer tissue(P<0.05)and is closely related to the patients' serum CEA expression level.However,it is not related to gender,age and clinical diagnosis(p>0.05).Conclusion:1.The expression of differentially expressed protein AGR2 in colorectal cancer issues is higher than that in adjacent tissues.2.The upregulation of AGR2 may be related to the occurrence,development,invasion and metastasis of colorectal cancer,and is a promoting factor for the occurrence and development of colorectal cancer in China.3.It is an indicator of the potential diagnosis,detection,treatment and prognosis of colorectal cancer.However its specific mechanism in the progression of colorectal cancer needs further study...
