Objective:The objective of this study was to evaluate the effects of tea polyphenols combining vitamin C on cognitive function and relative serum biochemical markers in patients with type 2 diabetes mellitus and mild cognitive impairment.Providing scientific basis for effectively improving the cognitive function of patients.Methods:1.The type 2 diabetes mellitus patients were recruited from the Department of Clinic Endocrinology of the First Hospital of Shanxi Medical University,we examined cognitive function by using Montreal Cognitive Assessment(MoCA).According to the criteria of mild cognitive impairment,inclusion and exclusion criteria,selected the appropriate patients.2.All the enrolled patients were divided into control group,tea polyphenols group,tea polyphenols combines vitamin C group or vitamin C group,respectively.All participants received the interventions of dietary,physical activities and the lifestyle.Tea polyphenols group was provided capsule of tea polyphenols(300mg/d),tea polyphenols combines vitamin C group was provided capsule of tea polyphenols(300mg/d)and vitamin C(300mg/d),respectively.Vitamin C group was given capsule of vitamin C(300mg/d),this trial lasted 6 months.Before and after the intervention,we tested cognitive function by using MoCA.Serum total protein,albumin,prealbumin,total cholesterol,triglyceride,High Density Lipoprotein-Cholesterol,Low Density Lipoprotein-Cholesterol,glycosylat-ed hemoglobin,fasting plasma glucose were all needed to be measured.Screening the relevant changes of inflammatory and neurotrophic factors,which includes 20 factors,by using AAH-NEU-1 kit.3.Data were conducted by SPSS software(20.0 SPSS,Inc,IL).Analysis of Variance,Chi-square test and Kruskal-Wallis test were used to compare the differences of baseline characteristics in different groups.We used multivariable regression analysis assessed whether tea polyphenols combines vitamin C can influence the cognitive function and the levels of serum total protein,albumin,prealbumin,glycosylated hemoglobin and fasting plasma glucose.Results:1.At baseline: A total of 79 subjects were enrolled in the study,20 patients(mean age was 63.75±5.68 y)in the control group,20(aged 66.40±7.54 y)in the tea polyphenols group,19(aged 61.26±6.63 y)in the tea polyphenols combines vitamin C group,20(aged 65.10±8.03 y)in the vitamin C group.There were no significant differences in baseline demographic and clinical biochemical markers(P>0.05)between treatment groups and control group,except sex and height(P<0.05).2.Cognitive function: After adjusting sex and serum lipids,intervention was the factor of MoCA scores.After 6 months of trial,the ? value and 95%CI in tea polyphenols combines vitamin C group was 4.078(2.064,6.093),the scores of MoCA were higher than control group.There were no significant differences in the scores of MoCA between the tea polyphenols group as well as vitamin C group and control group.3.Inflammatory factors and neurotrophic factors: After invention,there were 5 factors changed by using protein chip in control group,the levels of IL-10,VEGF-A,IL-8,TNF-? were up,however,the level of IL-1? was down.The levels of IL-10,VEGF-A,S100 B,?-NGF,IL-8,TNF-?,IL-1? were all up and the level of MCP-1 was down in tea polyphenols group.The levels of IL-10,VEGF-A,S100 B,?-NGF,IL-8 were down in the tea polyphenols combines vitamin C group.The levels of IL-10,S100 B,?-NGF,IL-8,IL-1?,MCP-1 and TNF-? were down,however,the level of VEGF-A was up in the vitamin C group.4.Serum glycosylated hemoglobin,fasting plasma glucose and proteins: After adjusting sex and serum lipids,intervention had no influence of glycosylated hemoglobin,total proteins,albumin and prealbumin.However,intervention was the factor of fasting plasma glucose.After 6 months of trial,the ? value and 95%CI in tea polyphenols group,tea polyphenols combines vitamin C group and vitamin C was-1.395(-2.514,-0.277),-1.450(-2.677,-0.198),-1.217(-2.350?-0.084),respectively.The levels of fasting plasma glucose were all lower than control group.Conclusion:1.Tea polyphenols combines vitamin C could improve the cognitive function in patients with type 2 diabetes mellitus and mild cognitive impairment.2.The levels of IL-10,VEGF-A,S100 B,?-NGF and IL-8 were down-regulated in tea polyphenols combines vitamin C.Tea polyphenols combines vitamin C can adjust the levels of inflammatory factors and neurotrophic factors.3.Tea polyphenols combines vitamin C could improve the levels of fasting plasma glucose,but there were no cooperative relationship in improving the levels of glycosylated hemoglobin,total protein,albumin and prealbumin.
