| Objective : Non-targeted Metabolomic of Banxia Xiexin Decoction in treating chronic atrophic gastritis by liquid Chromatography-Mass Spectrometry,screening out potential differential metabolites,to elucidate the molecular mechanism of Banxia Xiexin Decoction on chronic atrophic gastritis.Methods:(1)The chromatogram of total ion base peak was obtained by LC-MS technique,and then multivariate statistical PCA,PLS-DA,OPLS-DA analysis was carried out,the components satisfying P<0.05 and VIP>1 were selected as differential metabolites,(2)According to the ms/ms model,the fragment information was matched in the HMDB、Metlin、massbank 、Lipid Maps 、mzclound database to obtain the accurate metabolite information,(3)Correlation analysis of different metabolites among groups,the cor.test()function in R(v3.1.3)was used to test the correlation analysis of metabolites,further screening of differential metabolites with p-value ≤ 0.05,(4)The differential metabolites were enriched by metpa and the key metabolic pathways were screened out.Results:(1)Under the condition of this research analysis,the pretreated serum obtained16553,8409 molecules respectively,and the urine obtained 9287,6850 molecules respectively.(2)Through multivariate statistical analysis,24 serum samples were obtained from rats with chronic atrophic gastritis,10 differential metabolites were up-regulated and14 were down-regulated.the metabolic pathways involved are sphingomyolipid metabolism,ascorbic acid and almond metabolism,niacin and nicotinamide metabolism,histidine metabolism,lysine degradation,glutathione metabolism,glycerol metabolism,arachidonic acid metabolism,tryptophan metabolism,arginine and proline metabolism,aminoacyl-t RNA biosynthesis,purine metabolism,68 differential metabolites were obtained in urine,12 up-regulated and 56 down-regulated,The metabolic pathways involved are d-glutamine and d-glutamic acid metabolism,histidine metabolism,purine metabolism,nitrogen metabolism,tyrosine metabolism,arginine and proline metabolism,butyric acid metabolism,biotin metabolism,alanine metabolism,aspartic acid and glutamic acid metabolism,ascorbic acid and almond metabolism,nicotinic acid and nicotinamide metabolism,pentose and glucuronide conversion,pyrimidine metabolism,lysine degradation,citric acid cycle(TCA cycle),starch and sucrose metabolism,inositol phosphate metabolism,glutathione metabolism,porphyrin and chlorophyll metabolism,cysteine and methionine metabolism,glycine,serine and threonine metabolism,amino acyl-trna biosynthesis,tryptophan metabolism.(3)There were 23 differential metabolites,10up-regulated and 13 down-regulated,91 differential metabolites in urine,65 up-regulation and 26 down-regulation in CAG rats,which indicated that the main metabolic pathways were d-glutamine and d-glutamic acid metabolism,Linoleic acid metabolism,arginine and proline metabolism,nitrogen metabolism,pentose and glucuronic acid conversion,histidine metabolism,pentose phosphate pathway,alanine,aspartic acid and glutamic acid metabolism,cysteine and methionine metabolism,glycine-serine-threonine metabolism,pyrimidine metabolism,unsaturated fatty acid biosynthesis,aminoacyl-t RNA biosynthesis,purine metabolism,etc.(4)BXD ancient group were 32 serum differential metabolites,17up-regulation,15 down-regulation,104 urine differential metabolites,45 up-regulation and59 down-regulation in CAG rats.The results showed that the metabolic pathway was mainly composed of unsaturated fatty acid biosynthesis,panquinone and other terpenoid quinone biosynthesis,phenylalanine-tyrosine-tryptophan biosynthesis,vitamin metabolism,amino acyl-t RNA biosynthesis,arachidonic acid metabolism,phenylalanine metabolism,α-linolenic acid metabolism,Arginine and proline metabolism,histidine metabolism,β-alanine metabolism,lysine degradation,,glycolysis or glycosylation,glutathione metabolism,cysteine and methionine metabolism,tyrosine metabolism,purine metabolism,etc.(5)BXD Pharmacopoeia Group,44 serum differential metabolites were obtained,13up-regulated,31 down-regulated,107 urine differential metabolites,35 up-regulated and72 down-regulated.The main metabolic pathways involved were β-alanine metabolism and glutathione metabolism,and the metabolic pathways involved were β-alanine metabolism,glutathione metabolism,histidine metabolism,arginine and proline metabolism,alanine-aspartic-glutamic metabolism,d-glutamine and d-glutamic acid metabolism,linoleic acid metabolism,glycine-serine-threonine metabolism,nitrogen metabolism,arachidonic acid metabolism,Panthenic acid and Co A biosynthesis,glyoxylic acid and dicarboxylic acid metabolism,butyric acid metabolism,citric acid cycle(TCA cycle),pyruvate metabolism,aminoacyl-t RNA biosynthesis,purine metabolism,glycolic acid metabolism,pyrimidine metabolism,tryptophan metabolism,The biosynthesis of unsaturated fatty acids,the biosynthesis of primary bile acids and so on.Conclusion:(1)The metabolic pathways related to the pathological development of cag were analyzed by establishing a lc-ms-based non-targeted metabolomics method: purine metabolism,ascorbic acid-bitter almond metabolism,d-glutamine-d-glutamic acid metabolism,tyrosine metabolism,and so on.Histidine metabolism,arginine-proline metabolism,guanosine,guanine,inosine,adenine,dglucuronic acid,ketoglutaric acid,gentianic acid,dopamine,norepinephrine,vanillic acid,L-glutamic acid,L-histidine,L-arginine,4-acetamidobutanoic acid,It is possible to predict the diagnostic markers of CAG.(2)Effect of Banxia Xiexin Decoction on 8-chlortheophylline,urinary acid,hypervanillic acid,inosine,dtmp,guanine,L-glutamic acid,L-glutamic amide,(-)-stercobilin,d-galactose,cyclic guanosine phosphate,guanosine cyclase,adenine,spermine,spermidine,L-arginine,genkwanin,(r)-panacid,taurine,Scopoline and arachidonic acid have regulatory effects.(3)The mechanisms of Banxia Xiexin Decoction in treating CAG are nitrogen metabolism,purine metabolism,histidine metabolism,alanine-aspartic-glutamic metabolism,glutathione metabolism,aminoacyl-t RNA biosynthesis and tyrosine metabolism.The results of this study can be used as the most relevant metabolic pathway and metabolite reference for BXD in the treatment of CAG.In this study,the mechanism of Banxia Xiexin Decoction on CAG was found to be related to nucleotide metabolism,amino acid metabolism and glucose metabolism,and the specific manifestations were inhibition of inflammatory reaction in the stomach and inhibition of tumor necrosis factor.Improve the immune system,improve the defense ability of the body and so on. |
