Objectives:This paper quantitatively analyzed the Ziyuglycoside and tannins before and after processing of Sanguisorba officinalis,and determined their coagulative activity.It explained the material basis for the hemostasis of Sanguisorba officinalis by stirring charcoal,which provided scientific basis for optimizing the processing technology of Sanguisorba officinalis carbon and improving its quality standard Methods:Uv-vis spectrophotometry was used to determine the total tannin content in the carbon of Sanguisorba officinalis with different processing degrees.The contents of Gallic acid and Ellagic acid in Sanguisorba officinalis carbon were analyzed by UPLC.HPLC method was used to analyze the changes of saponins in radix ulmaculata thunb.In vitro coagulation assay was used to determine the coagulant activity of different degree of processing Sanguisorba officinalis charcoal.Gray correlation analysis method was used to analyze the correlation between the carbon fingerprint data and coagulation index of Sanguisorba officinalis to find the material basis related to coagulation activity.Results:Through the study on the components of Ziyuglycoside I,Ziyuglycoside II,Gallic acid,Ellagic acid and total tannin in the charcoal of Sanguisorba officinalis with different processing degrees,the regularity of chemical composition changes after frying of elm was found.The components of Gallic acid and Ellagic acid of tannin monomer were significantly increased after Sanguisorba officinalis was processed into Sanguisorba officinalis carbon.According to the correlation analysis results of in vitro coagulation and fingerprint chromatographic peaks,there were 36 chromatographic peaks with a correlation degree of > 0.6 and 10 chromatographic peaks with a correlation degree of > 0.7,but no chromatographic peaks had a correlation degree of > 0.8 with coagulation.The peak of FIB content associated chromatography overlaps with that of in vitro coagulation test.Peaks 6,7,14 and 28 are common associated peaks of two pharmacodynamic indexes.No.14 peak accounted for the highest proportion of peak area,and it was the component with the highest content in the five chromatographic peaks with the highest correlation with the content of FIB in Sanguisorba officinalis carbon.The component was identified as Gallic acid,which indicated that it was an important component in Sanguisorba officinalis charcoal for coagulation and hemostasis.After processing,the correlation of Ellagic acid with significantly increased content of components on in vitro coagulation activity was only 0.4712,and that with FIB containing lead was only 0.4441,indicating that the correlation of this component on coagulation activity was weak.Conclusions:Through the quantitative and qualitative analysis of the components before and after processing and combining with the results of clotting experiment and clotting experiment,this paper analyzed the components related to clotting effect and preliminarily explained the material basis of clotting effect of Sanguisorba officinalis carbon.The conclusion of this paper provides scientific basis for optimizing the processing technology and improving the quality standard of Sanguisorba officinalis carbon,which is of great significance for improving the quality of prepared slices.
