Objectives:The chemical composition of Sanguisorba officinalis was isolated and studied to obtain the active ingredients or indicators of pharmacological effects,which provided basic support for the interpretation of the physical basis of pharmacological actions such as hemostatic,and also provided support for quality evaluation research.Established the TLC,HPLC fingerprint and simultaneous determination the contents of Ziyuglycoside ? and Ziyuglycoside ?,in order to improve the quality standard of Sanguisorba officinalis,and ensure clinical efficacy.Methods:The ethanol extracts of Sanguisorba officinalis was extracted with n-butyl alcohol,dichloromethane and ethyl acetate.The extracts of ethyl acetate were seprated and purificated by using silica gel column chromatography,sephadex chromatography method,recrystallization and semi-preparative high performace liquid chromatograph and so on.The obtained compounds were analyzed and identified according to physical and chemical properties,NMR,ESI-MS,and the spectral analysis data was compared with the relevant references.Based on the systematic isolation and analysis of chemical composition of Sanguisorba officinalis,quantitative analysis with a single maker(QAMS)was employed with the saponins and tannins as the quality evaluation indexes.A scientific and rational quality evaluation method was established by TLC identification and HPLC characteristic profiling method and other quality evaluation methods,and analyzed the statistics to develop the limit of content as the quality evaluation of Sanguisorba officinalis.Results:From the extracts of ethyl acetate we seperated and identified 4 chemical compounds,which were identified as Gallic acid(?),Ethyl gallate(?),Ziyuglycoside?(?),Ziyuglycoside?(?).The isolated Ziyuglycoside?and Ziyuglycoside?were used as reference substance to establish a method for the determination of saponins content in Sanguisorba officinalis,the method had good repeatability and is simple and feasible.The content of Ziyuglycoside ? and Ziyuglycoside ? in 10 batches of Sanguisorba officinalis in 6 provinces was determined.According to the statistic analysis of the data,the content limit of Ziyuglycoside?was suggested to be no less than 3.7%,the content limit of Ziyuglycoside?was suggested to be no less than 0.21%.The similarity degree of HPLC fingerprint of Sanguisorba officinalis was greater than 0.90,twenty one mutual peaks were selected in chromatography,and the most of the detected peaks were separated effectively.Conclusions:Isolated compounds from Sanguisorba officinalis and identified tiheir stuctures.A method was established to analyze the content of Ziyuglycoside?and Ziyuglycoside ? in Sanguisorba officinalis,improved quality standard of Sanguisorba officinalis,saponins could be used as one of the quality control indexes.Established the HPLC fingerprint of Sanguisorba officinalis,The similarity between 10 batches of Sanguisorba officinalis was greater than 0.90,showing that the quality of 10 batches of Sanguisorba officinalis was consistent.The method was simple and feasible and could be used to effectively control the quality of Sanguisorba officinalis.
