Abjective:A mouse model of unilateral ureteral obstruction(UUO)was used to investigate whether Qigui Yishen decoction has effect of reducing the renal inflammatory response and renal fibrosis by affecting coagulation and fibrinolysis.Methods:40 ICR mice were divided into five groups randomly,including Qigui Yishen group(10 g/kg/d),Benazepril group(10 mg/kg/d),Ligustrazine group(100mg/kg/d),model group and the sham group.All groups were used for UUO model except the shamgroup.Each group of mice was continuously intragastrically administered the respective drugs for 9 days.On the 10th day,mice were sacrificed by cervical dislocation.The renal tissue was observed by HE and Masson staining,and the renal interstitial damage was observed.Imnunohistochemistry and Western blot were used to detect the expression of transforming growth factor-beta 1(TGF-?1)?Plasminogen activator inhibitor type-1(PAI-1)?thrombomodulin(TM)?fibronectin(FN)in the renal tissue.Results:Compared with the sham group,the renal interstitial fibrosis index of the model group,the Qigui Yishen group,the Benazepril group and the Ligustrazine group increased significantly.The expression of TGF-beta 1?PAI-1?FN protein increased obviously,and the expression of TM decreased significantly(P<0.05).Compared with the model group,the Qigui Yishen group,the Benazepril group and the Ligustrazine group had obvious effect of decreasing renal interstitial fibrosis and expression of TGF-beta 1?PAI-1?and FN,while increasing expression of TM.The Qigui Yishen group was superior to the Benazepril group and the Ligustrazine group(P<0.05).Conclusion:Qigui Yishen decoction has a therapeutic effect on delaying renal fibrosis in UUO model mice.The mechanism is related to reducing the expression of PAI-1 in the renal tissue and increasing the expression of TM in the renal tissue or reducing its metabolism.
