Objective: Exploring the prognostic factors of advanced non-small cell lung cancer (NSCLC) to offer reference data for the individual treatment plan .Methods: 155 cases of advanced NSCLC, from September 2004 to May 2010 treated in the Forth Hospital of He Bei Medical University, diagnosed by cytopathology or histopathology , were analysed in gender, age, athology type, smoke, target treatment, the number of lot chemotherapy, therapy method, prognostic index and the single nucleotide polymorphisms (SNPs) of MMP-12 and MMP-13 genes,whose clinical document were collected and the SNPs of MMP-12 and MMP-13 genes were detected. All the cases were follow-up visited for 63 months, and the last time for visition was in December, 2010. Statistical analysis was performed using SPSS13.0 software package (SPSS Company, Chicago, Illinois, USA). Kaplan–Meier method(Product Limit method)was used to calculate the survival rate, its significance was tested by log-rank test, factors related to the prognosis were analyzed by the method of Cox Regression Analysis. The statistical result is significante if the P<0.05.Results:1 The rate of follow-up visition was 100%. The overall median survival time was 12.00 monthes, 1-year survival rate was 46.20%, 2-year survival rate was 23.40%, 3-year survival rate was 13.20%.2 In the single factor analysis, the age (≤65years, >65years; P=0.018), targeted therapy (with, without; P=0.008), therapy method (chemotherapy plus radiotherapy, chemotherapy, radiotherapy, supportive treatment; P=0.000), the number of lot chemotherapy (from 0 to 8 cycles; P=0.001) and prognostic index (≤1, >1; P=0.003) had the statistics significance (P<0.05), while the gender and the SNPs of MMP-12 and MMP-13 genes had not statistics significance(P>0.05). 3 In multi-factor analysis, the number of lot chemotherapy (P=0.001), athology type (P=0.010), smoke (P=0.013) and prognostic index (P=0.004) had statistics significance (P<0.05).Conclusion:1 The number of lot chemotherapy and prognostic index were important factors affecting prignosis of advanced NSCLC.2 The age, athology type, target treatment, smoke and therapy method had some effect on advanced NSCLC, but could not as the independent-prognosis factor.3 The gender and the SNPs of MMP-12 and MMP-13 genes had not the statistics significance in this investigation.
