Research On Quality Standard Of Standard Decoction Of White Chrysanthemum Of Hangzhou

During development on the modernization of traditional Chinese medicine,the problem that how to standardize the utilization of drug should be solved.Decoction is an important link,because it isthe most widely used medicine in clinical application.Standard decoction is just a standard substance that can measure the drug use form of numbers of decoction pieces and products produced by different enterprises in a standardized way.It can be used to control drug quality as a whole,and connect decoction pieces,herbal extracts,decoction and other drug forms of utilization.The chrysanthemum,as the common clinical medicinal material,has the efficacy of expelling wind,clearing heat,calming the liver,improving eyesight and detoxifying,besides it is indicated by the modern pharmacological researches that chrysanthemum has anti-inflammation,antimicrobial,antineoplastic,antioxidant and other functions.Currently,eight varieties of commodity chrysanthemum are under mixed used in clinical application.At present,white chrysanthemum of Hangzhou is the variety with largest output and dosage in the medicinal chrysanthemum market.In this experiment,10 batches of medicinal material produced by the genuine producing areas and main producing areas of white chrysanthemum of Hangzhou were adopted.The preparation method of standard decoction of white chrysanthemum of Hangzhou is set up.The main procedures including Adding water 12 times the amount of medicinal material to the medicinal material,fully soaking under the standing state for 30min,heat to the slight boiling,to conduct reflux for 30min,filtering with a soaked eight layer gauze while it is hot,adding water 10 times the amount of the medicinal material to the residue once more,continue to conduct reflux for 20 min,filtering with the same method,combining the two filtrates,and conducting vacuum concentration under 60? to 200mL,namely,the metered volume is that each milliliter of water contains solution equivalent to 0.05 g decoction pieces.The quality evaluation method of standard decoction of white chrysanthemum of Hangzhou was explored and established,and such method was established from character,pH value,ratio of extraction,relative density,transfering rate and fingerprint.Character of the standard decoction of chrysanthemum of Hangzhou prepared by the standard preparation method is the light fawn clear water solution.and some of them w ere slightly yellow green,with the relative density betw een 1.007 and 1.011 pH value ware 5.37 to 5.56.Average ratio of extraction was 23.6%,range from 19.93%to 29.69%.transfering rate was 56.2%when calculated as per chlorogenic acid,scope of which was from 51.8%to 63.9%.Galuteolin was 57.4%on average,scope of which was from 51.9%to 64.4%.3.5-O-dicaffeoyl-quinic acid was 30.6%on average,and the scope of which was from 21.7%to 38.5%,and the similarity of chromatographic fingerprint was 0.864?0.989.The standard decoction of white chrysanthemum of Hangzhou was applied to the evaluation of Chinese medicinal granule.Two kinds of commercially available chrysanthemum formula granules were evaluated according to the standard of standard decoction of chrysanthemum morifolium established.Characteristic of Sample 2 was identical to that of the standard decoction of white chrysanthemum of Hangzhou,while color of Sample 1 was relatively deeper,with opaque solution.Relative density and pH value were not quite different from the standard decoction of white chrysanthemum of Hangzhou.Upon content determination,it was found that compareing with the standard decoction,the content of chlorogenic acid of Sample 1 was within the scope,meeting the standard of the Pharmacopoeia when converted to crude drug,content of other relative constituents were extremely low.The content of galuteolin and 3,5-O-dicaffeoyl-quinic acid were even not detected at all.The content of all constituents of Sample 2 formula granule were within the content scope value of standard decoction.However,after being converted to the equivalent crude drug weight according to the indication,the content of decoction pieces obtained was much lower than the standard indicated by the Chinese Pharmacopoeia(2015 Edition)for chrysanthemum,and the similarity of Sample 1 was only 0.164.Meanwhile,the similarity of Sample 2 was 0.826.With this quality evaluation method,quality of standard decoction is examined in an all-round way,indicating that quality of the standard decoction obtained is reliable and representative,and can be used for evaluating other standard drug use forms.
